In this protocol we describe an in vitro assay to study the 3D migrational behavior of cells in matrix environments that topologically resemble ECMs encountered in vivo. There are three main strengths of this assay as compared to other currently available methods. First, this assay allows one to simultaneously examine the cell migration mechanisms at both population level and individual cell level. This opens up possibilities of studying collective cell migration13, which has to date been largely limited to 2D assays14, while at the same time observing the behavior of the individual cell within the population. For cells that do not naturally migrate in groups or clusters, the assay offers a way to analyze the migratory properties in detail from a large number of cells with diverse local microenvironment, which can provide important insights into the biophysical origins of migration efficiency and directional persistence7,15.
Second, by modifying the gel conditions of the inner and outer gels, one can directly study the effect of matrix mechanics and microstructure on 3D cell migration behavior10. This is especially important, as it is known that cell migration and cancer invasion are highly influenced by matrix mechanics and porosity16,17, and that soft tissues in our body have a very diverse topology18. Moreover, the method is readily adaptable for matrices other than collagen. This is advantageous, as not all cell lines can spread and migrate in collagen-based matrices. A critical step that must be noted is that, to ensure physical connectivity of the gel interface, the formation of the outer gel must be initiated before complete polymerization of the inner gel. Since this is strongly dependent on the specific polymerization kinetics of the gels (also for collagen prepared under different conditions, e.g., concentration, polymerization pH, temperature, cross-linking8), an optimal polymerization duration should first be systematically identified. Imperfections in the interfacial connectivity between the inner and outer gel usually results in a gap and collection of the spreading cell population in the interface. Note, however, that perfect interfacial connectivity can introduce a practical problem: the boundary between the inner and outer gels becomes less obvious when imaging. If necessary, sparse fiducial markers, such as inert beads, can be used to distinguish the inner and outer gels.
Third, the possibility of long-term 3D culture (up to 3 weeks in our laboratory) and live-cell imaging allows a wide application of the assay. It is now possible to obtain spatially-resolved viscoelastic measurements of the local matrices surrounding the cells using microrheological approaches19. Furthermore, our recent study10 reveals that the efficacy of various anti-migratory drugs (administered on the 7th day of the experiment) can be strongly affected by the architecture and stiffness of the matrix (i.e., outer gel). In the future, we plan to use these experimental frameworks to further understand how such effects can arise from the basic molecular mechanisms of cell migration (e.g., cytoskeletal contractility20, extracellular matrix reorganization21, and pericellular proteolysis22).
One limitation of this assay is the asymmetry of the mechanical support: the top surface of the gel is not attached to any solid surface, while the bottom surface adheres to the glass surface of the dish. While edge effects can be avoided simply by excluding cells near the edges, the lack of mechanical support at the top surface can potentially lead to in-plane preferential migration and therefore introduce artificial flattening of the 3D cell trajectories7. One way to solve this issue while still maintaining oxygen and nutrition supply to the cells is by topping the gel with holed glass plate23. Additionally, to further suppress any wall effect, a thin layer of collagen gel can be first made on the glass surfaces, before the formation of the inner and outer gels. This ensures complete geometrical encapsulation of the gels.
To obtain meaningful and accurate description of cell migration behavior, it is important to consider the time scale and the nature of migration, which can vary considerably between cell lines. Preliminary experiments must be systematically performed to understand these factors, and then the interval time between image captures and live-cell imaging duration can be adjusted. The interval time should be no larger than the characteristic time scale of cell turning. It is also important to consider that cell migration behavior may change both spatially and temporally, depending on the biophysical conditions of the local microenvironment7. It is always a good idea to simultaneously visualize the cells and the fibrous matrix, for example using multi-color confocal fluorescence or multimodal (e.g., reflectance, second harmonic generation) imaging. After imaging, post-processing of the time-lapse 3D image, construction of the cell tracks, and analysis of the trajectories can in principle be done with any image processing software.
In conclusion, the concentric gel assay described here provides a simple and useful experimental system for quantitatively studying 3D cell migration, mechanosensing and mechanotransduction in diverse microenvironments.

Migration von Zellen spielt eine wichtige Rolle bei verschiedenen physiologischen Reaktionen, wie der Embryonalentwicklung, Hämostase und Immunantwort als auch in pathologischen Prozessen, wie Gefäßerkrankungen, Entzündungen und Krebs 1. Sezieren der biochemischen und biophysikalischen Faktoren Zellmigration ist es daher von grundlegender Bedeutung nicht nur für die Grundprinzipien der zellulären Funktionen zu verstehen, sondern auch für verschiedene biomedizinische Anwendungen, wie zB im Tissue Engineering, Anti-Metastasen und entzündungshemmenden Medikamentenentwicklung voranzutreiben. Da in vivo Beobachtung ist technisch anspruchsvoll, hat viele Bemühungen auf die in vitro Rekapitulation Zellmigration konzentriert. In-vitro-Verfahren zur Zellmigration zu untersuchen sind weitgehend für Assays auf zweidimensionale (2D) Oberflächen der Grund auf neu konzipiert worden, vor allem oder Wundheilungs Test 2. Solche Tests bieten einfachen Versuchsaufbau, einfache Live-Cell Imaging und liefern nützliche Einblicke in verschiedene biochemische Mechanismen der Zellmigration zugrunde liegen. Allerdings sind diese Tests nicht für die extrazelluläre Matrix (ECM) Architektur und Umbau, die kritischen Aspekte im Verständnis in vivo Migration berücksichtigen. Kürzlich wurde zunehmend klar, dass ein 3D-Kulturmodell häufig Kollagen-Matrices auf der Basis 3, eine Plattform, die besser ähnelt der in vivo Situation. Tatsächlich Zellen zeigen Migrations Dynamik, die sich von denen auf 2D Oberflächen sind, insbesondere aufgrund der unterschiedlichen Dimensionalität der Umgebung 4. Darüber hinaus sind die biophysikalischen und mechanischen Eigenschaften der Matrix beeinflussen empfindlich Zellmigration 5, auch im Rahmen der Tumorzellinvasion 6. Hier präsentieren wir eine Methode zur 3D-Zellmigrationsverhalten in ECM mit biophysikalischen Eigenschaften, die mit Herstellungsbedingungen leicht variiert werden kann studieren. Die Zellen sindin einem &quot;inneren Gel&quot; ausgesät und dürfen sich zu entkommen und dringen in die anfänglich zellfreie &quot;äußeren Gel&quot;. Das Verfahren stützt sich auf die Fähigkeit der Zelle, das Vorhandensein von zu erkennen, und die Neigung in der äußeren Gel, das in engem Zusammenhang mit Zell mechanosensing 7 verbunden ist, in zellfreien Bereiche migrieren. In dieser Studie beschäftigen wir Kollagen Netzwerke wie die ECM von hoch invasiven Brustkrebszellen, MD-MBA-231 eingedrungen. Die mechanischen Eigenschaften und die Mikrostruktur sowohl der inneren und äußeren Gele können abgestimmt werden, 8 und 9 dadurch um physiologisch relevante Bedingungen zu erreichen. Rekonstruktion und Analyse der Zellspuren erlauben detaillierte quantitative Untersuchung der raumzeitlichen Wanderungsverhalten sowohl auf Bevölkerungsebene und Einzelzellebene. Wichtig ist, dass der Aufbau des konzentrischen Gelsystem ahmt die in vivo Gewebetopologie durch wandernde Zellen, insbesondere Krebszellen eindringen und bietet somit wichtige Einblicke in konfrontiertan die physikalischen Mechanismen der Zellmigration und Metastasierung.

concentric gel system to study the biophysical role of matrix microenvironment on 3d cell migration

Die Fähigkeit von Zellen zur Migration entscheidend ist in einer Vielzahl von Zellfunktionen während des gesamten Lebens aus embryonalen Entwicklung und bei der Wundheilung zur Tumor- und Krebsmetastasierung. Trotz intensiver Forschung, die grundlegenden biochemischen und biophysikalischen Prinzipien der Zellwanderung immer noch nicht vollständig verstanden, insbesondere in ihre physiologisch relevanten dreidimensionalen (3D) Mikroumgebungen. Hier beschreiben wir ein in vitro Test zur quantitativen Untersuchung der 3D-Zellmigration Verhalten ermöglichen. Das Verfahren nutzt mechanosensing Fähigkeit der Zelle, und die Neigung, in vorher unbesetzten extrazellulären Matrix (ECM) zu migrieren. Wir nutzen die Invasion von hochinvasiven Brustkrebs-Zellen, MDA-MB-231, in Kollagengele als Modellsystem. Die Verbreitung der Zellpopulation und die Migrationsdynamik einzelner Zellen über Wochen Kultur kann mit Abbildung lebender Zellen überwacht und analysiert, um raumzeitlich aufgelösten Daten zu extrahieren. Darueber HinausDas Verfahren ist für verschiedene extrazelluläre Matrizen leicht an und bietet so eine einfache, aber leistungsstarke Möglichkeit, die Rolle der biophysikalischen Faktoren in der Mikroumgebung auf die Zellmigration zu untersuchen.

Die konzentrische Geltest hier vorgestellt wurde unter Verwendung hochinvasiven Brustkrebszellen MDA-MB-231 mit 2,4 mg / ml inneren Kollagen-Gel und einer Zellansiedlungsdichte = 2 x 10 6 Zellen / ml, als Beispiel. Wie in Abbildung 2 dargestellt, in der Regel nach wenigen Tagen Kultur verletzt die Zellen die Innen-Außen-Gel-Grenzfläche und damit begonnen, die äußeren Gel einzudringen. Die Zellpopulation verbreiten überwiegend radial nach außen. Die Polymerisationsbedingungen der äußeren Gel kann modifiziert werden, um die Rolle der Dichte und die mechanischen Eigenschaften der Matrix auf die Zellmigration Eigenschaften zu untersuchen, Fig. 3 zeigt die Bewegungen der einzelnen Zellen in der äußeren Gelen von 1,5 mg / ml, 2,4 mg / ml, und 4,0 mg / ml Kollagen, polymerisiert bei pH 7,4. Konfokale Zeitrafferbilder wurden an der Grenze zwischen dem inneren und äußeren Gel für 8 h erfasst und die Verschiebung der Zellen in den drei verschiedenen Kollagen Konzenrationen in weißen Pfeile angedeutet. Abhängig von der Zellansiedlungsdichte und Proliferationsrate, ~ 200 Einzelzelle Trajektorien typischerweise extrahiert und in jeder Probe analysiert. Wir quantitativ analysiert die Zellbahnen in den 3 verschiedenen Kollagenkonzentrationen bezogen auf die durchschnittliche Netto-Verschiebung, zurückgelegte Strecke, Richtungs Ausdauer und mittlere Geschwindigkeit über 8 h der Bildgebung (Abbildung 4). Es wurde beobachtet, dass die mittlere Verschiebung und Entfernung höchsten waren für 4,0 mg / ml-Konzentration auf 58 &amp; mgr; m und 141,5 &amp; mgr; m, jeweils, allerdings gab es keinen signifikanten Unterschied in der Verschiebung und der Abstand zwischen den Zellen in 2,4 mg / ml und 4,0 mg / ml Gel . In 1,5 mg / ml Gel, die mittlere Verschiebung und Abstand kleinsten waren. Diese Beobachtung scheint zu den jüngsten Berichten, die zeigen, dass die Invasion Effizienz sinkt in Gelen mit zunehmender Kollagenkonzentration 11,12 widersprechen. Man beachte jedoch, dass die Verschiebungen undEntfernungen in unserem Test werden aus allen Einzelzellen Spuren innerhalb überwiegend radial nach außen wandernden Bevölkerung gemessen. Diese Parameter sind daher viel weniger anfällig für die schnellsten laufende Zellsubpopulation und nicht vollständig durch stochastische Bewegungen dominiert. In allen Fällen ist die Gesamt abgedeckt (4B) Abstand größer als die Nettoverschiebung (4A). Persistenz definieren einfach als das Verhältnis zwischen der Verschiebung der Entfernung, der erhalten wir Persistenz von ~ 0,4 in allen Gel-Bedingungen (4C). Diese relativ geringe Persistenz spiegelt die eigen schwache gerichtete Migration der Zellen in unseren Assays. Wir erwarten, dass das Vorhandensein von chemotaktischen Stimuli Richtungs oder biochemischen Gradienten wird die Persistenz potentiell in Kollagen konzentrationsabhängigen Weise erhöht. Darüber hinaus beobachteten wir auch, dass die mittlere Geschwindigkeit der Zellmigration nicht signifikant wi ändernth Kollagenkonzentration (4D), wahrscheinlich aufgrund der Wechselwirkung zwischen Matrix Steifigkeit (das mit Kollagen-Konzentration erhöht) und die Porengröße 10 sowie der Raumzeit-Veränderung der Migrationseigenschaften innerhalb der Zellpopulation 7 (das mit Kollagen-Konzentration abnimmt). Die Durchschnittsgeschwindigkeit zwischen 31 &amp; mgr; m / h und 37,5 &amp; mgr; m / h variiert wird, mit einem Minimum einzelnen Zellgeschwindigkeit von 7 &amp; mgr; m / h und eine maximale Einzelzellengeschwindigkeit von 114 um / h. <img alt="Abbildung 1" src="/files/ftp_upload/52735/52735fig1.jpg" /> Abbildung 1. Design of a Cyclic Pressure Bioreactor for the Ex Vivo Study of Aortic Heart Valves

The aortic valve, located between the left ventricle and the aorta, allows for unidirectional blood flow, preventing backflow into the ventricle. Aortic valve leaflets are composed of interstitial cells suspended within an extracellular matrix (ECM) and are lined with an endothelial cell monolayer. The valve withstands a harsh, dynamic environment and is constantly exposed to shear, flexion, tension, and compression. Research has shown calcific lesions in diseased valves occur in areas of high mechanical stress as a result of endothelial disruption or interstitial matrix damage1-3. Hence, it is not surprising that epidemiological studies have shown high blood pressure to be a leading risk factor in the onset of aortic valve disease4. 
The only treatment option currently available for valve disease is surgical replacement of the diseased valve with a bioprosthetic or mechanical valve5. Improved understanding of valve biology in response to physical stresses would help elucidate the mechanisms of valve pathogenesis. In turn, this could help in the development of non-invasive therapies such as pharmaceutical intervention or prevention. Several bioreactors have been previously developed to study the mechanobiology of native or engineered heart valves6-9. Pulsatile bioreactors have also been developed to study a range of tissues including cartilage10, bone11 and bladder12. The aim of this work was to develop a cyclic pressure system that could be used to elucidate the biological response of aortic valve leaflets to increased pressure loads. 
The system consisted of an acrylic chamber in which to place samples and produce cyclic pressure, viton diaphragm solenoid valves to control the timing of the pressure cycle, and a computer to control electrical devices. The pressure was monitored using a pressure transducer, and the signal was conditioned using a load cell conditioner. A LabVIEW program regulated the pressure using an analog device to pump compressed air into the system at the appropriate rate. The system mimicked the dynamic transvalvular pressure levels associated with the aortic valve; a saw tooth wave produced a gradual increase in pressure, typical of the transvalvular pressure gradient that is present across the valve during diastole, followed by a sharp pressure drop depicting valve opening in systole. The LabVIEW program allowed users to control the magnitude and frequency of cyclic pressure. The system was able to subject tissue samples to physiological and pathological pressure conditions. This device can be used to increase our understanding of how heart valves respond to changes in the local mechanical environment.

Study of Cell Migration in Microfabricated Channels

The method described here allows the study of cell migration under confinement in one dimension. It is based on the use of microfabricated channels, which impose a polarized phenotype to cells by physical constraints. Once inside channels, cells have only two possibilities: move forward or backward. This simplified migration in which directionality is restricted facilitates the automatic tracking of cells and the extraction of quantitative parameters to describe cell movement. These parameters include cell velocity, changes in direction, and pauses during motion. Microchannels are also compatible with the use of fluorescent markers and are therefore suitable to study localization of intracellular organelles and structures during cell migration at high resolution. Finally, the surface of the channels can be functionalized with different substrates, allowing the control of the adhesive properties of the channels or the study of haptotaxis. In summary, the system here described is intended to analyze the migration of large cell numbers in conditions in which both the geometry and the biochemical nature of the environment are controlled, facilitating the normalization and reproducibility of independent experiments.

Analysis of Cell Migration within a Three-dimensional Collagen Matrix

The ability to migrate is a hallmark of various cell types and plays a crucial role in several physiological processes, including&#160;embryonic development, wound healing, and immune responses. However, cell migration is also a key mechanism in cancer enabling these cancer cells to detach from the primary tumor to start metastatic spreading. Within the past years various cell migration assays have been developed to analyze the migratory behavior of different cell types. Because the locomotory behavior of cells markedly differs between a two-dimensional (2D) and three-dimensional (3D) environment it can be assumed that the analysis of the migration of cells that are embedded within a 3D environment would yield in more significant cell migration data. The advantage of the described 3D collagen matrix migration assay is that cells are embedded within a physiological 3D network of collagen fibers representing the major component of the extracellular matrix. Due to time-lapse video microscopy real cell migration is measured allowing the determination of several migration parameters as well as their alterations in response to pro-migratory factors or inhibitors. Various cell types could be analyzed using this technique, including lymphocytes/leukocytes, stem cells, and tumor cells. Likewise, also cell clusters or spheroids could be embedded within the collagen matrix concomitant with analysis of the emigration of single cells from the cell cluster/ spheroid into the collagen lattice. We conclude that the 3D collagen matrix migration assay is a versatile method to analyze the migration of cells within a physiological-like 3D environment.

Planar Gradient Diffusion System to Investigate Chemotaxis in a 3D Collagen Matrix

The importance of cell migration can be seen through the development of human life. When cells migrate, they generate forces and transfer these forces to their surrounding area, leading to cell movement and migration. In order to understand the mechanisms that can alter and/or affect cell migration, one can study these forces. In theory, understanding the fundamental mechanisms and forces underlying cell migration holds the promise of effective approaches for treating diseases and promoting cellular transplantation. Unfortunately, modern chemotaxis chambers that have been developed are usually restricted to two dimensions (2D) and have complex diffusion gradients that make the experiment difficult to interpret. To this end, we have developed, and describe in this paper, a direct-viewing chamber for chemotaxis studies, which allows one to overcome modern chemotaxis chamber obstacles able to measure cell forces and specific concentration within the chamber in a 3D environment to study cell 3D migration. More compelling, this approach allows one to successfully model diffusion through 3D collagen matrices and calculate the coefficient of diffusion of a chemoattractant through multiple different concentrations of collagen, while keeping the system simple and user friendly for traction force microscopy (TFM) and digital volume correlation (DVC) analysis.

Cell migration is an important part of human development and life. Preparation of 3D Collagen Gels and Microchannels for the Study of 3D Interactions In Vivo

Historically, most cellular processes have been studied in only 2 dimensions. While these studies have been informative about general cell signaling mechanisms, they neglect important cellular cues received from the structural and mechanical properties of the local microenvironment and extracellular matrix (ECM). To understand how cells interact within a physiological ECM, it is important to study them in the context of 3 dimensional assays. Cell migration, cell differentiation, and cell proliferation are only a few processes that have been shown to be impacted by local changes in the mechanical properties of a 3-dimensional ECM. Collagen I, a core fibrillar component of the ECM, is more than a simple structural element of a tissue. Under normal conditions, mechanical cues from the collagen network direct morphogenesis and maintain cellular structures. In diseased microenvironments, such as the tumor microenvironment, the collagen network is often dramatically remodeled, demonstrating altered composition, enhanced deposition and altered fiber organization. In breast cancer, the degree of fiber alignment is important, as an increase in aligned fibers perpendicular to the tumor boundary has been correlated to poorer patient prognosis1. Aligned collagen matrices result in increased dissemination of tumor cells via persistent migration2,3. The following is a simple protocol for embedding cells within a 3-dimensional, fibrillar collagen hydrogel. This protocol is readily adaptable to many platforms, and can reproducibly generate both aligned and random collagen matrices for investigation of cell migration, cell division, and other cellular processes in a tunable, 3-dimensional, physiological microenvironment.

Tunable Hydrogels from Pulmonary Extracellular Matrix for 3D Cell Culture

Here we present a method for establishing multiple component cell culture hydrogels for in vitro lung cell culture. Beginning with healthy en bloc lung tissue from porcine, rat, or mouse, the tissue is perfused and submerged in subsequent chemical detergents to remove the cellular debris. Histological comparison of the tissue before and after processing confirms removal of over 95% of double stranded DNA and alpha galactosidase staining suggests the majority of cellular debris is removed. After decellularization, the tissue is lyophilized and then cryomilled into a powder. The matrix powder is digested for 48 hr in an acidic pepsin digestion solution and then neutralized to form the pregel solution. Gelation of the pregel solution can be induced by incubation at 37 &#176;C and can be used immediately following neutralization or stored at 4 &#176;C for up to two weeks. Coatings can be formed using the pregel solution on a non-treated plate for cell attachment. Cells can be suspended in the pregel prior to self-assembly to achieve a 3D culture, plated on the surface of a formed gel from which the cells can migrate through the scaffold, or plated on the coatings. Alterations to the strategy presented can impact gelation temperature, strength, or protein fragment sizes. Beyond hydrogel formation, the hydrogel stiffness may be increased using genipin.

Measuring Global Cellular Matrix Metalloproteinase and Metabolic Activity in 3D Hydrogels

Three-dimensional (3D) cell culture systems often more closely recapitulate in vivo cellular responses and functions than traditional two-dimensional (2D) culture systems. However, measurement of cell function in 3D culture is often more challenging. Many biological assays require retrieval of cellular material which can be difficult in 3D cultures. One way to address this challenge is to develop new materials that enable measurement of cell function within the material. Here, a method is presented for measurement of cellular matrix metalloproteinase (MMP) activity in 3D hydrogels in a 96-well format. In this system, a poly(ethylene glycol) (PEG) hydrogel is functionalized with a fluorogenic MMP cleavable sensor. Cellular MMP activity is proportional to fluorescence intensity and can be measured with a standard microplate reader. Miniaturization of this assay to a 96-well format reduced the time required for experimental set up by 50% and reagent usage by 80% per condition as compared to the previous 24-well version of the assay. This assay is also compatible with other measurements of cellular function. For example, a metabolic activity assay is demonstrated here, which can be conducted simultaneously with MMP activity measurements within the same hydrogel. The assay is demonstrated with human melanoma cells encapsulated across a range of cell seeding densities to determine the appropriate encapsulation density for the working range of the assay. After 24 h of cell encapsulation, MMP and metabolic activity readouts were proportional to cell seeding density. While the assay is demonstrated here with one fluorogenic degradable substrate, the assay and methodology could be adapted for a wide variety of hydrogel systems and other fluorescent sensors. Such an assay provides a practical, efficient and easily accessible 3D culturing platform for a wide variety of applications.

A Human 3D Extracellular Matrix-Adipocyte Culture Model for Studying Matrix-Cell Metabolic Crosstalk

The extracellular matrix (ECM) plays a central role in regulating tissue homeostasis, engaging in crosstalk with cells and regulating multiple aspects of cellular function. The ECM plays a particularly important role in adipose tissue function in obesity, and alterations in adipose tissue ECM deposition and composition are associated with metabolic disease in mice and humans. Tractable in vitro models that permit dissection of the roles of the ECM and cells in contributing to global tissue phenotype are sparse. We describe a novel 3D in vitro model of human ECM-adipocyte culture that permits study of the specific roles of the ECM and adipocytes in regulating adipose tissue metabolic phenotype. Human adipose tissue is decellularized to isolate ECM, which is subsequently repopulated with preadipocytes that are then differentiated within the ECM into mature adipocytes. This method creates ECM-adipocyte constructs that are metabolically active and retain characteristics of the tissues and patients from which they are derived. We have used this system to demonstrate disease-specific ECM-adipocyte crosstalk in human adipose tissue. This culture model provides a tool for dissecting the roles of the ECM and adipocytes in contributing to global adipose tissue metabolic phenotype and permits study of the role of the ECM in regulating adipose tissue homeostasis.

Finite Element Modelling of a Cellular Electric Microenvironment

Clinical studies show electrical stimulation (ES) to be a potential therapy for the healing and regeneration of various tissues. Understanding the mechanisms of cell response when exposed to electrical fields can therefore guide the optimization of clinical applications. In vitro experiments aim to help uncover those, offering the advantage of wider input and output ranges that can be ethically and effectively assessed. However, the advancements in in vitro experiments are difficult to reproduce directly in clinical settings. Mainly, that is because the ES devices used in vitro differ significantly from the ones suitable for patient use, and the path from the electrodes to the targeted cells is different. Translating the in vitro results into in vivo procedures is therefore not straightforward. We emphasize that the cellular microenvironment's structure and physical properties play a determining role in the actual experimental testing conditions and suggest that measures of charge distribution can be used to bridge the gap between in vitro and in vivo. Considering this, we show how in silico finite element modelling (FEM) can be used to describe the cellular microenvironment and the changes generated by electric field (EF) exposure. We highlight how the EF couples with geometric structure to determine charge distribution. We then show the impact of time dependent inputs on charge movement. Finally, we demonstrate the relevance of our new in silico model methodology using two case studies: (i) in vitro fibrous Poly(3,4-ethylenedioxythiophene) poly(styrenesulfonate) (PEDOT-PSS) scaffolds and (ii) in vivo collagen in extracellular matrix (ECM).

Stepwise Cell Seeding on Tessellated Scaffolds to Study Sprouting Blood Vessels

The cardiovascular system is a key player in human physiology, providing nourishment to most tissues in the body; vessels are present in different sizes, structures, phenotypes, and performance depending on each specific perfused tissue. The field of tissue engineering, which aims to repair or replace damaged or missing body tissues, relies on controlled angiogenesis to create a proper vascularization within the engineered tissues. Without a vascular system, thick engineered constructs cannot be sufficiently nourished, which may result in cell death, poor engraftment, and ultimately failure. Thus, understanding and controlling the behavior of engineered blood vessels is an outstanding challenge in the field. This work presents a high-throughput system that allows for the creation of organized and repeatable vessel networks for studying vessel behavior in a 3D scaffold environment. This two-step seeding protocol shows that vessels within the system react to the scaffold topography, presenting distinctive sprouting behaviors depending on the compartment geometry in which the vessels reside. The obtained results and understanding from this high throughput system can be applied in order to inform better 3D bioprinted scaffold construct designs, wherein fabrication of various 3D geometries cannot be rapidly assessed when using 3D printing as the basis for cellularized biological environments. Furthermore, the understanding from this high throughput system may be utilized for the improvement of rapid drug screening, the rapid development of co-cultures models, and the investigation of mechanical stimuli on blood vessel formation to deepen the knowledge of the vascular system.

Engineered tissues heavily rely on proper vascular networks to provide vital nutrients and gases and remove metabolic waste. In this work, a stepwise seeding protocol of endothelial cells and support cells creates highly organized vascular networks in a high-throughput platform for studying developing vessel behavior in a controlled 3D environment.
