Transgenic manipulation of subsets of brain cells is increasingly used for studying behaviors and their underlying neural circuits. In Drosophila, the GAL4-upstream activating sequence (UAS) binary system is powerful for gene manipulation, but GAL4 expression is often too broad for fine mapping of neural circuits. Here, we describe the development of unique molecular genetic tools to restrict GAL4 expression patterns. Building on the GAL4-UAS system, our method adds two components: a collection of enhancer-trap recombinase, Flippase (ET-FLP), transgenic lines that provide inheritable, reproducible, and tissue-specific FLP and an FRT-dependent GAL80 "flip-in" construct that converts FLP expression into tissue-specific repression of GAL4 by GAL80. By including a UAS-encoded fluorescent protein, circuit morphology can be simultaneously marked while the circuit function is assessed using another UAS transgene. In a proof-of-principle analysis, we applied this ET-FLP-induced intersectional GAL80/GAL4 repression (FINGR) method to map the neural circuitry underlying fly wing inflation. The FINGR system is versatile and powerful in combination with the vast collection of GAL4 lines for neural circuit mapping as well as for clonal analysis based on the infusion of the yeast-derived FRT/FLP system of mitotic recombination into Drosophila. The strategies and tactics underlying our FINGR system are also applicable to other genetically amenable organisms in which transgenes including the GAL4, UAS, GAL80, and FLP factors can be applied.

A genetic mosaic approach for neural circuit mapping in Drosophila.

La manipulation transg&#xE9;nique de sous-ensembles de cellules du cerveau est de plus en plus utilis&#xE9; pour l&#x26;#39;&#xE9;tude des comportements et de leurs sous-jacents circuits neuronaux. Chez la drosophile, le GAL4-s&#xE9;quence d&#x26;#39;activation en amont du syst&#xE8;me binaire (UAS) est puissant pour la manipulation g&#xE9;n&#xE9;tique, mais GAL4 expression est souvent trop large pour la cartographie fine des circuits neuronaux. Ici, nous d&#xE9;crivons le d&#xE9;veloppement de la uniques outils de g&#xE9;n&#xE9;tique mol&#xE9;culaire pour restreindre GAL4 profils d&#x26;#39;expression. Une approche g&#xE9;n&#xE9;tique pour la cartographie mosa&#xEF;que des circuits neuronaux chez la drosophile

Transgene Manipulation von Teilmengen von Gehirnzellen wird zunehmend verwendet, f&uuml;r die Untersuchung der Verhaltensweisen und ihre zugrunde liegenden neuronalen Schaltkreise. Drosophila das GAL4-upstream aktivierende Sequenz (FH)-Bin&auml;r-System ist m&auml;chtig f&uuml;r Genmanipulation, aber GAL4 Ausdruck ist oft zu breit f&uuml;r Feinkartierung von neuronale Schaltkreise. Das FINGR-System ist vielseitig und leistungsf&auml;hig in Kombination mit der riesigen Sammlung von GAL4-Linien f&uuml;r die neuralen Schaltkreis Zuordnung sowie f&uuml;r klonale Analyse auf der Grundlage der Infusion des Hefe-abgeleitete FRT/FLP-Systems Mitotische Rekombination in Drosophila. Die Strategien und Taktiken, die zugrunde liegenden unser FINGR-System gelten auch f&uuml;r andere genetisch empf&auml;nglich Organismen, in denen, die transgene einschlie&szlig;lich die GAL4, UAS, GAL80 und FLP Faktoren angewendet werden kann.

La manipulaci&#xF3;n transg&#xE9;nica de subconjuntos de c&#xE9;lulas del cerebro se utilizan cada vez m&#xE1;s para el estudio de los comportamientos y sus circuitos neurales subyacentes. En Drosophila, la GAL4-aguas arriba secuencia de activaci&#xF3;n (UAS) sistema binario es de gran alcance para la manipulaci&#xF3;n gen&#xE9;tica, pero la expresi&#xF3;n de GAL4 es demasiado amplio para el mapeo preciso de los circuitos neuronales. En este caso, se describe el desarrollo de herramientas &#xFA;nicas de gen&#xE9;tica molecular para restringir los patrones de expresi&#xF3;n GAL4. Bas&#xE1;ndose en el sistema GAL4-UAS, nuestro m&#xE9;todo se agregan dos componentes: una recopilaci&#xF3;n de potenciador de la trampa de la recombinasa, Flippase (ET-FLP), las l&#xED;neas transg&#xE9;nicas que proporcionan FLP reproducibles heredable, y espec&#xED;fico de tejido y un GAL80 FRT-dependiente &#x26;quot;flip en &#x26;quot;construir que convierte la expresi&#xF3;n de FLP en tejidos espec&#xED;ficos de la represi&#xF3;n de GAL4 por GAL80.
