荧光与过氧化物的反应在 HRP 的存在下产生高荧光卤是建立了良好的48,49。HRP 催化氧化荧光荧光至荧光卤红50,51。这种氧化作用是由反应 (OH) 和 HDL 脂质过氧化物 (LOOH) 中的内源性过氧化物所驱动的。没有胆固醇氧化酶, 卤 (HRP) 可以量化的内在高密度脂蛋白脂质过氧化物含量的特定数量的高密度脂蛋白胆固醇。从每个井的荧光读数中减去缓冲器的空气氧化后产生的 OH 的背景产量。卤在大多数样品中具有最小的自发荧光。在反应缓冲液中加入过氧化氢酶 (1-4 U/毫升) 可以迅速抑制内源性过氧化物, 从而使荧光读出时间的增加是由高密度脂质过氧化物驱动的。该方法具有通用性, 能评估高密度脂蛋白胆固醇酯与游离高密度脂蛋白脂质过氧化物含量的比值32,48,49 。
有许多变化的荧光 HDL 功能检测32。简要地有至少3主要方法: a) 增加每井的特定容量 hdl (例如50µL) 和以后规范化 hdl 脂质过氧化值由水平的 hdl C 如确定在临床实验室;b) 根据标准比色胆固醇测定法确定每个样品中高密度脂蛋白 c 的浓度, 然后在每井中添加一特定量的高密度脂蛋白 c (如1µg);和 c) 通过 hdl 或》 I 蛋白含量32使 hdl 功能读数正常化。还有至少3主要的方法, 如何隔离 hdl 的高密度脂蛋白脂质过氧化测定: a) 高密度脂蛋白胆固醇沉淀, 如与聚乙二醇;b) 亲和捕获;和 c) 用于高密度脂蛋白隔离的其他标准非高通量方法, 如µLtracentrifugation32。此外, 该化验是多才多艺的, 可以评估脂质过氧化物含量的高密度脂蛋白胆固醇酯与游离高密度脂蛋白32。有几种方法可以报告结果, 例如, 任意荧光单位、标准化的卤荧光单元、归一化比率到汇集控件32。在此, 我们提出了最具重现性的方法。
如果使用等离子, 那么在含有柠檬酸钠的试管中制备血浆是很重要的, 如抗凝血27,28。EDTA40和肝素硫酸根可干扰氧化反应41,42。肝素硫酸盐可干扰 HDL 功能的生化测定27,28。虽然冷冻保存的血清和血浆对血脂、hdl 功能和 hdl 脂质过氧化的测定是次优, 但冷冻保存的标本可以量化 hdl 脂质过氧化的相对差异。重要的是要处理在一个特定的研究中的所有样本完全相同的方式 (如相同的冻融循环), 并包括在每个板块的汇集控制, 以解释潜在的混杂与不同的研究之间的样品处理差异。长期冷冻可能会损害 hdl 功能测定结果43但是, 如果使用了一个适当的池控制27, 则在一项研究中的样本间 hdl 脂质过氧化的相对差异仍可评估, 28。
重要的是, 先前的 HDL 功能检测没有标准化。在这里, 我们描述一种方法, 使用适当的控制确保读数的标准化。更具体地说, 有几个已知的参数, 可能会影响的读出 HDL 功能的生化检测, 如基质效应 (血清 vs 方法的等离子体制备, 存在的白蛋白和其他蛋白质), 冷冻保存, 冻融32. 高密度脂蛋白功能荧光测定可以使用商用标准和实验试剂32进行标准化。然而, 也有在每项研究中, 在不同的人中, 可能影响 HDL 功能结果测定的几个未知 (或特征) 混杂。因此, 在每一个板块中, 我们使用一个从特定的样本感兴趣的具体的研究 (已处理相同的) 的混合高密度脂蛋白控制, 最大限度地减少工件和混杂32。使用血浆血库样本和高密度脂蛋白 C 值从临床实验室可以进一步标准化化验32。
我们以前已经表明, 不同的高密度脂蛋白隔离方法可以显着影响的读数生化检测 hdl 功能27,28,32但荧光可以可靠地测量 HDLox 无论的高密度脂蛋白隔离32。高密度脂蛋白分离方法是高密度脂蛋白功能研究的主要限制因素。离心今天被认为是引用方法, 但仍然是一个耗时的方法52。电泳不精确, 不规范53。高密度脂蛋白沉淀方法涉及使用 polyanions, 如聚乙二醇 (PEG) 沉淀低密度脂蛋白离开高密度脂蛋白在上清54。虽然这些方法显示某些缺点例如可变的结果与 lipemic 血清和干涉与酵素胆固醇规程55对原始的做法的早先修改使用规范化的商业可利用试剂产生一个简单、可靠和准确的过程56。虽然 PEG 沉淀是常用的方法分离 hdl21,57从患者血清, 一个主要问题是存在的非高密度脂蛋白蛋白, 如白蛋白58。亲和隔离可用于减少白蛋白对 HDL 功能的影响32。虽然可以评估特定捕获方法的 HDLox 的相对差异, 但针对 hdl 的特定抗体可能无法完全捕获复杂的高密度脂蛋白结构。采用本协议所述的适当控制, 可以可靠地确定高密度脂蛋白标本 (通过 PEG 沉淀分离) HDLox 之间的相对差异。
这种方法与其他所有 HDL 功能的测定一样, 都有其关键的局限性。在体内动脉壁上 HDL 的氧化是相当复杂的, 异质性和脂质过氧化物含量可能只部分反映 HDLox46。高密度脂蛋白结构和功能不断改变在体内和测量的 hdl 功能在一个 timepoint 可能无法反映的影响, hdl 功能障碍的最终器官疾病的时间59。不知道 HDLox 读数是否应该由高密度脂蛋白 C 或高密度脂蛋白蛋白22来正常化。未来的临床研究应该验证的重要性, 检测读数 (hdl 脂质过氧化物含量每毫克的高密度脂蛋白 C)。
总之, 荧光 hdl 功能测定是一种可靠的方法, 以确定高密度脂蛋白脂质过氧化物含量每特定数量的高密度脂蛋白 (HDLox)。较高的 HDLox 值与降低的 HDL 抗氧化功能相关。该方法的读数与经验证的细胞检测, 心血管疾病的替代措施, 全身炎症, 免疫功能障碍和相关的心血管和代谢风险表型19, 21 ,27,32,343536373847. 此方法为检测高密度脂蛋白功能在人类疾病中的作用提供了一个方便而有力的工具。

动脉粥样硬化性心血管疾病 (CVD) 是导致全球死亡的主要原因1,2。流行病学研究表明, 低浓度高密度脂蛋白 (HDL) 胆固醇通常与动脉粥样硬化发展的风险成反比1,2。虽然几项研究支持高密度脂蛋白1,2的 atheroprotective 作用, hdl 减轻动脉粥样硬化的起始和进展的机制是复杂的3,4。因此, 有人建议, HDL 的复杂结构和功能, 而不是绝对水平可以更准确地预测动脉粥样硬化5,6,7,8。一些高密度脂蛋白蛋白和脂质成分的改变会损害 hdl 的功能, 如动脉粥样硬化等炎症状态。这些 i) 降低其胆固醇流出电位9, ii) 减少消炎和增加 HDL 相关的炎蛋白6,7, iii) 降低抗氧化因子水平和活性和 HDLs抑制低密度脂蛋白氧化的能力 (LDLox)10和 iv) 增加脂质过氧化氢含量和氧化还原活性 (HDLox)9,11。对 hdl (如胆固醇外流、抗氧化功能) 的 pleotropic 功能进行评估的健壮性分析可以补充临床上 hdl-高密度脂蛋白 C 的测定。
HDL 功能通常由细胞方法评估, 如胆固醇流出试验8,12,13,14。这些方法有很大的局限性, 包括对所使用的细胞类型、报告的读数类型、缺乏标准化和甘油三酯的混杂效应7,15。这些缺点给大型临床研究带来困难16。与细胞检测相比, 无单元检测可能给 HDL 功能提供更有力的措施。胆固醇外流是 HDL 最重要的功能之一, 但它只能通过细胞检测来确定。其他方法来确定 HDL 功能, 如蛋白质组学17,18,19,20,21,22,23, 24和细胞单核蛋白的趋化性分析 HDL 功能17,22,25尚未标准化, 不能用于大规模的人类研究。
HDL 具有显著的抗氧化剂 atheroprotective 效果5,6,7,8。HDL 的抗氧化功能已确定在 LDL 的存在前细胞游离荧光分析26。这些生化荧光方法的高密度脂蛋白抗氧化功能最初开发的穆罕默德 Navab 和艾伦 Fogelman 及其同事26。虽然许多人类研究使用这些方法来确定 HDL 功能 17,18,19,20,21,22,23 ,24, 脂质 (高密度脂蛋白) 脂质 (LDL) 和脂质荧光相互作用可能会限制这些细胞游离酶生化检测 HDL 功能的重复性27,28。

A Fluorescence-based Assay of Phospholipid Scramblase Activity

Scramblases translocate phospholipids across the membrane bilayer bidirectionally in an ATP-independent manner. The first scramblase to be identified and biochemically verified was opsin, the apoprotein of the photoreceptor rhodopsin. Rhodopsin is a G protein-coupled receptor localized in rod photoreceptor disc membranes of the retina where it is responsible for the perception of light. Rhodopsin&#39;s scramblase activity does not depend on its ligand 11-cis-retinal, i.e., the apoprotein opsin is also active as a scramblase. Although constitutive and regulated phospholipid scrambling play an important role in cell physiology, only a few phospholipid scramblases have been identified so far besides opsin. Here we describe a fluorescence-based assay of opsin&#39;s scramblase activity. Opsin is reconstituted into large unilamellar liposomes composed of phosphatidylcholine, phosphatidylglycerol and a trace quantity of fluorescent NBD-labeled PC (1-palmitoyl-2-{6-[7-nitro-2-1,3-benzoxadiazole-4-yl)amino]hexanoyl}-sn-glycero-3-phosphocholine). Scramblase activity is determined by measuring the extent to which NBD-PC molecules located in the inner leaflet of the vesicle are able to access the outer leaflet where their fluorescence is chemically eliminated by a reducing agent that cannot cross the membrane. The methods we describe have general applicability and can be used to identify and characterize scramblase activities of other membrane proteins.

A Simple and Rapid Protocol for Measuring Neutral Lipids in Algal Cells Using Fluorescence

Algae are considered excellent candidates for renewable fuel sources due to their natural lipid storage capabilities. Robust monitoring of algal fermentation processes and screening for new oil-rich strains requires a fast and reliable protocol for determination of intracellular lipid content. Current practices rely largely on gravimetric methods to determine oil content, techniques developed decades ago that are time consuming and require large sample volumes. In this paper, Nile Red, a fluorescent dye that has been used to identify the presence of lipid bodies in numerous types of organisms, is incorporated into a simple, fast, and reliable protocol for measuring the neutral lipid content of Auxenochlorella protothecoides, a green alga. The method uses ethanol, a relatively mild solvent, to permeabilize the cell membrane before staining and a 96 well micro-plate to increase sample capacity during fluorescence intensity measurements. It has been designed with the specific application of monitoring bioprocess performance. Previously dried samples or live samples from a growing culture can be used in the assay.

A simple protocol to determine the neutral lipid content of algal cells using a Nile Red staining procedure is described. This time-saving technique offers an alternative to traditional gravimetric-based lipid quantification protocols. It has been designed for the specific application of monitoring bioprocess performance.

A High-Throughput Luciferase Assay to Evaluate Proteolysis of the Single-Turnover Protease PCSK9

Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a single-turnover protease which regulates serum low-density lipoprotein (LDL) levels and, consequently, cardiovascular disease. Although PCSK9 proteolysis is required for its full hypercholesterolemic effect, the evaluation of its proteolytic function is challenging: PCSK9 is only known to cleave itself, undergoes only a single turnover, and after proteolysis, retains its substrate in its active site as an auto-inhibitor. The methods presented here describe an assay which overcomes these challenges. The assay focuses on intermolecular proteolysis in a cell-based context and links successful cleavage to the secreted luciferase activity, which can be easily read out in the conditioned medium. Via sequential steps of mutagenesis, transient transfection, and a luciferase readout, the assay can probe PCSK9 proteolysis under conditions of either genetic or molecular perturbation in a high-throughput manner. This system is well suited for both the biochemical evaluation of clinically discovered missense single-nucleotide polymorphisms (SNPs), as well as for the screening of small-molecule inhibitors of PCSK9 proteolysis.

This protocol presents a method to evaluate the proteolytic activity of an intrinsically low-activity, single turnover protease in a cellular context. Specifically, this method is applied to evaluate the proteolytic activity of PCSK9, a key driver of lipid metabolism whose proteolytic activity is required for its ultimate hypercholesterolemic function.

Enrichment of Native Lipoprotein Particles with microRNA and Subsequent Determination of Their Absolute/Relative microRNA Content and Their Cellular Transfer Rate

Lipoprotein particles are predominately transporters of lipids and cholesterol in the bloodstream. Furthermore, they contain small amounts of strands of noncoding microRNA (miRNA). In general, miRNA alters the protein expression profile due to interactions with messenger-RNA (mRNA). Thus, knowledge of the relative and absolute miRNA content of lipoprotein particles is essential to estimate the biological effect of cellular particle uptake. Here, a quantitative real-time polymerase chain reaction (qPCR)-based protocol is presented to determine the absolute miRNA content of lipoprotein particles&#8212;exemplified shown for native and miRNA-enriched lipoprotein particles. The relative miRNA content is quantified using multiwell microfluidic array cards. Furthermore, this protocol allows scientists to estimate the cellular miRNA and, thus, the lipoprotein particle uptake rate. A significant increase of the cellular miRNA level is observable when using high-density lipoprotein (HDL) particles artificially loaded with miRNA, whereas incubation with native HDL particles yields no significant effect due to their rather low miRNA content. In contrast, the cellular uptake of low-density lipoprotein (LDL) particles&#8212;neither with native miRNA nor artificially loaded with it&#8212;did not alter the cellular miRNA level.

Here, a quantitative real-time polymerase chain reaction-based protocol is presented for the determination of the native micro-RNA content (absolute/relative) of lipoprotein particles. In addition, a method for increasing the micro-RNA level, as well as a method for determining the cellular uptake rate of lipoprotein particles, is demonstrated.

Click-Chemistry Based Fluorometric Assay for Apolipoprotein N-acyltransferase from Enzyme Characterization to High-Throughput Screening

Lipoproteins from proteobacteria are posttranslationally modified by fatty acids derived from membrane phospholipids by the action of three integral membrane enzymes, resulting in triacylated proteins. The first step in the lipoprotein modification pathway involves the transfer of a diacylglyceryl group from phosphatidylglycerol onto the prolipoprotein, resulting in diacylglyceryl prolipoprotein. In the second step, the signal peptide of prolipoprotein is cleaved, forming an apolipoprotein, which in turn is modified by a third fatty acid derived from a phospholipid. This last step is catalyzed by apolipoprotein N-acyltransferase (Lnt). The lipoprotein modification pathway is essential in most &#947;-proteobacteria, making it a potential target for the development of novel antibacterial agents. Described here is a sensitive assay for Lnt that is compatible with high-throughput screening of small inhibitory molecules. The enzyme and substrates are membrane-embedded molecules; therefore, the development of an in vitro test is not straightforward. This includes the purification of the active enzyme in the presence of detergent, the availability of alkyne-phospholipids and diacylglyceryl peptide substrates, and the reaction conditions in mixed micelles. Furthermore, in order to use the activity test in a high-throughput screening (HTS) setup, direct readout of the reaction product is preferred over coupled enzymatic reactions. In this fluorometric enzyme assay, the alkyne-triacylated peptide product is rendered fluorescent through a click-chemistry reaction and detected in a multiwell plate format. This method is applicable to other acyltransferases that use fatty acid-containing substrates, including phospholipids and acyl-CoA.

Presented here is a sensitive fluorescence assay to monitor apolipoprotein N-acyltransferase activity using diacylglyceryl peptide and alkyne-phospholipids as substrates with click-chemistry.

Fluorescence-Based Measurements of Phosphatidylserine/Phosphatidylinositol 4-Phosphate Exchange Between Membranes

Several members of the evolutionarily conserved oxysterol-binding protein (OSBP)-related proteins(ORP)/OSBP homologs (Osh) family have recently been found to represent a novel lipid transfer protein (LTP) group in yeast and human cells. They transfer phosphatidylserine (PS) from the endoplasmic reticulum (ER) to the plasma membrane (PM) via PS/phosphatidylinositol 4-phosphate (PI(4)P) exchange cycles. This finding allows a better understanding of how PS, which is critical for signaling processes, is distributed throughout the cell and the investigation of the link between this process and phosphoinositide (PIP) metabolism. The development of new fluorescence-based protocols has been instrumental in the discovery and characterization of this new cellular mechanism in vitro at the molecular level. This paper describes the production and the use of two fluorescently labelled lipid sensors, NBD-C2Lact and NBD-PHFAPP, to measure the ability of a protein to extract PS or PI(4)P and to transfer these lipids between artificial membranes. First, the protocol describes how to produce, label, and obtain high-purity samples of these two constructs. Secondly, this paper explains how to use these sensors with a fluorescence microplate reader to determine whether a protein can extract PS or PI(4)P from liposomes, using Osh6p as a case study. Finally, this protocol shows how to accurately measure the kinetics of PS/PI(4)P exchange between liposomes of defined lipid composition and to determine lipid transfer rates by fluorescence resonance energy transfer (FRET) using a standard fluorometer.

A Quantitative Fluorescence Microscopy-based Single Liposome Assay for Detecting the Compositional Inhomogeneity Between Individual Liposomes

Most research employing liposomes as membrane model systems or drug delivery carriers relies on bulk read-out techniques and thus intrinsically assumes all liposomes of the ensemble to be identical. However, new experimental platforms able to observe liposomes at the single-particle level have made it possible to perform highly sophisticated and quantitative studies on protein-membrane interactions or drug carrier properties on individual liposomes, thus avoiding errors from ensemble averaging. Here we present a protocol for preparing, detecting, and analyzing single liposomes using a fluorescence-based microscopy assay, facilitating such single-particle measurements. The setup allows for imaging individual liposomes in a massive parallel manner and is employed to reveal intra-sample size and compositional inhomogeneities. Additionally, the protocol describes the advantages of studying liposomes at the single liposome level, the limitations of the assay, and the important features to be considered when modifying it to study other research questions.

Lipid Exchange Assay in Living Cells

Lipid rafts are dynamic, ordered domains in the plasma membrane often formed during membrane protein clustering and signaling. The lipid identity of the outer leaflet drives the membrane&#39;s propensity to form lipid rafts. The transient nature of lipid rafts makes it difficult to study in living cells. Therefore, methods that add or remove raft-forming lipids at&#160;the outer leaflet of living cells facilitate studying the characteristics of rafts, such as their effects on membrane proteins. Lipid exchange experiments developed in our lab utilize lipid-loaded cyclodextrins to remove and add exogenous phospholipids to change the lipid constitution of the plasma membrane. Substituting the membrane with a raft or non-raft-forming lipid can aid in studying the effects on transmembrane protein activity. Here, we describe a method for lipid exchange on the outer leaflet of the plasma membrane using lipid-loaded cyclodextrin. We demonstrate the preparation of the exchange media and the subsequent treatment of attached mammalian cells. We also showcase how to measure the efficiency of exchange using HP-TLC. This protocol yields a nearly complete replacement of the outer leaflet with exogenous lipids without altering cellular viability, permitting further experimentation on modified intact plasma membranes.

High-throughput Nitrobenzoxadiazole-labeled Cholesterol Efflux Assay

Atherosclerosis leads to cardiovascular disease (CVD). It is still unclear whether cholesterol-HDL (cHDL) concentration plays a causal role in atherosclerosis development. However, an important factor in early stages of atheroma plaque formation is cholesterol efflux capacity to HDL (the ability of HDL particles to accept cholesterol from macrophages) in order to avoid foam cell formation. This is a key step in avoiding the accumulation of cholesterol in the endothelium and a part of reverse cholesterol transport (RCT) to eliminate cholesterol through the liver. Cholesterol efflux capacity to serum or plasma in macrophage cell models is a promising tool that can be used as biomarker for atherosclerosis. Traditionally, [3H]-cholesterol has been used in cholesterol efflux assays. In this study, we aim to develop a safer and faster strategy using fluorescent labelled-cholesterol (NBD-cholesterol) in a cellular assay to trace the cholesterol uptake and efflux process in THP-1-derived macrophages. Finally, we optimize and standardize the NBD-cholesterol efflux method and develop a high-throughput analysis using 96-well plates.

LDL Cholesterol Uptake Assay Using Live Cell Imaging Analysis with Cell Health Monitoring

The regulation of LDL cholesterol uptake through LDLR-mediated endocytosis is an important area of study in various major pathologies including metabolic disorder, cardiovascular disease, and kidney disease. Currently, there is no available method to assess LDL uptake while simultaneously monitoring for health of the cells. The current study presents a protocol, using a live cell imaging analysis system, to acquire serial measurements of LDL influx with concurrent monitoring for cell health. This novel technique is tested in three human cell lines (hepatic, renal tubular epithelial, and coronary artery endothelial cells) over a four-hour time course. Moreover, the sensitivity of this technique is validated with well-known LDL uptake inhibitors, Dynasore and recombinant PCSK9 protein, as well as by an LDL uptake promoter, Simvastatin. Taken together, this method provides a medium-to-high throughput platform for simultaneously screening pharmacological activity as well as monitoring of cell morphology, hence cytotoxicity of compounds regulating LDL influx. The analysis can be used with different imaging systems and analytical software.

This protocol provides an efficient approach to measuring LDL cholesterol uptake with real time influx rates using a live cell imaging system in various cell types. This technique provides a platform to screen the pharmacological activity of compounds affecting LDL influx while monitoring for cell morphology and hence potential cytotoxicity.
