AC16ヒト心筋細胞株は、BLUEFBIO(上海、中国)から購入したSV40形質転換ヒト線維芽細胞17と成体心室心臓組織からの初代細胞の融合に由来した。細胞株は、心筋細胞の特徴を多くの生化学的および形態学的特徴を開発する。また、細胞株は、ビトロ18,19において心筋損傷および心筋機能を評価するために広く用いられている。
1. 細胞培養
注:基底培養培地は、無血清のダルベッコの修飾されたイーグルの培地(DMEM)、10%のウシ胎児血清(FBS)、1%心臓筋細胞増殖サプリメント、および1%ペニシリン/ストレプトマイシン溶液で構成されています。使用前に4°C、前温37°Cで保存してください。
<ol><li>凍結保存されたヒト心筋細胞(HCM)細胞を液体窒素から取り出し、37°Cの水浴で解凍します。</li>
	<li>バイアルに小さな氷が残るまで、37°Cの水浴でバイアル(<1分)を軽く振ります。</li>
	<li>バイアルを無菌層流れフードに移します。70%のアルコールを浸した綿のボールでバイアルの外側を拭きます。</li>
	<li>解凍した細胞を含む遠心分離管に滴下する4mLの前温成長培地を移す。</li>
	<li>200×gで細胞懸濁液を10分間遠心分離する。 遠心分離後、上清を捨て、ペレットを完全培地の5mLに再懸濁します。</li>
	<li>95%の空気と5%のCO2を有する雰囲気の下で加湿インキュベーターで37°Cの細胞を維持する。 注:実験のために細胞を収穫する前に、細胞は約60〜70%の合流に達するまで増殖することができます。</li>
</ol>酸素グルコース欠乏(OGD)モデルの確立
注:研究期間の2時間前に、成長培地を無血清培地に置き換え、細胞を5%CO2の雰囲気下で37°Cで2時間加湿インキュベーターで再インキュベーターに再インキュベーターした。
<ol><li>培地を6ウェルプレートから吸引し、リン酸緩衝生理食塩液(PBS)で細胞を3回軽く洗浄します。</li>
	<li>1ウェルあたり2mLの新鮮な無糖DMEMを加えます。</li>
	<li>3ガスインキュベーターで、3ガスインキュベーターで、35%N2、5%CO2、および0.1%O2を37°Cで1、2、4、8、12、または16時間の一定温度で培養します。</li>
	<li>低酸素処理後、培地を6ウェルプレートから吸引し、PBS(pH7.4)で細胞を3回洗浄する。</li>
	<li>井戸ごとに完全なDMEMの2 mLを追加します。</li>
	<li>95%の空気と5%のCO2を有する雰囲気の下で加湿インキュベーターで37°Cの細胞を維持する。</li>
</ol>3. 時間温度プロトコル
注: 標準の時間温度プロトコルは、心臓手術中に使用されます。以下のプロトコルに従ってHCMを処理する(図1):タイムポイント1(T1)は誘導の終了を示し、タイムポイント2(T2)はメンテナンスの終了を示し、タイムポイント3(T3)は再温暖化の終了を示す。連続性標準条件下で維持された制御細胞を解析(37°C)。温度条件は、正確な温度調節を可能にする三ガスインキュベーターを使用して作成されます。
<ol><li>実験の2時間前に、培養液を6ウェルプレートから吸引し、1ウェルあたり2mLの新鮮な無血清DMEMを加える。</li>
	<li>5%CO2の雰囲気下で37°Cで2時間加湿インキュベーターで細胞を再インキュベートする。</li>
	<li>2時間後、培地を6ウェルプレートから吸引し、PBS(pH7.4)で細胞を3回洗浄する。</li>
	<li>1ウェルあたり2mLの新鮮な無血清DMEMを加えます。</li>
	<li>三ガスインキュベーター内の細胞を再インキュベートします。 メモ:取り付けられていない細胞や破片を取り除くために、媒体を交換してください。</li>
	<li>培養皿を三ガスインキュベーターに置くことにより、直ちに温度を変えます。</li>
	<li>1時間の冷却後、6ウェルプレートから培地を素早く吸引し、1ウェルあたり2mLの新鮮な無糖DMEMを加える。</li>
	<li>低酸素症を確立するために37°Cで95%N2、5%CO2、および0.1%O2を含む雰囲気下で三ガスインキュベーターで細胞を培養する。</li>
	<li>温度を以下のように設定します。 注:プロトコルは、低温処理の10時間から始まり、2時間から37°Cまでの再温暖化相、ノルメロザミア(37°C)の24時間の再温暖化が続きます。各時点で、3つのペトリ皿が分析のために取り除かれます。</li>
	<li>低温処理後、培地を6ウェルプレートから吸引し、PBS(pH7.4)で3回洗浄する。</li>
	<li>井戸ごとに完全なDMEMの2 mLを追加します。</li>
	<li>95%の空気と5%のCO2を有する雰囲気の下で加湿インキュベーターで37°Cの細胞を維持する。</li>
</ol>4. CCK-8実行可能性アッセイ
<ol><li>0.25%トリプシン-エチレンアミン酢酸(EDTA)溶液とPBSを使用前に37°Cに温めます。</li>
	<li>培地を吸引し、1 mLのPBSで細胞をすすい、ウェルの壁に沿って0.25%のトリプシンの0.5 mLを加えます。ほぼすべてのHCMが剥離されるまで37°Cでインキュベート(約1分)</li>
	<li>トリプシンを中和するためにウェルにDMEM完全培地の1 mLを追加します。</li>
	<li>細胞懸濁液を15mL遠心チューブに移し、500×gで遠心分離してHCMを3分間ペレットします。ペレットを邪魔することなく上清を吸引する。</li>
	<li>細胞懸濁液(5000細胞/ウェル)の100μLアリコートを96ウェルプレートに分配します。加湿インキュベーターでプレートを24時間前にインキュベートする(37°C)</li>
	<li>培養した細胞を96ウェルプレートに使用して、異なる処理群を生成します。</li>
	<li>インキュベーターでプレートを適当な時間(16時間)インキュベートします。</li>
	<li>プレートの各ウェルに10 μLのCCK-8溶液を加えます。</li>
	<li>インキュベーターでプレートを1時間インキュベートします。</li>
	<li>マイクロプレートリーダーを使用して450 nmで吸光度を測定します。 注意: 気泡は、ODの読み取り測定を妨げるので、井戸に気泡を導入しないように注意してください。</li>
</ol>5. アポトーシス解析のためのフローサイトメトリー
<ol><li>ステップ 4.2~4.4 に従って、トリプシン化と遠心化の手順を実行します。 メモ:細胞はEDTAなしでトリプシンで収穫されます。アポトーシスを評価するために、浮遊細胞と付着細胞の両方が収集される。</li>
	<li>1000×gで5分間細胞を遠心分離する。上清を捨て、PBSの1mLでペレットを再懸濁します。</li>
	<li>ヘモサイトメーターを使用して細胞を数えます。ピペットを使用して、トリパンブルー処理細胞懸濁液の100 μLをヘモサイトメーターに移します。ハンド集計カウンターを使用して、16個の正方形の1組に生きた未染色細胞を数え、PBSを使用して1×107 セル/mlで細胞懸濁液を生成する。</li>
	<li>200μLの細胞懸濁液(5×105-1×106細胞)を得る。 </li>
	<li>遠心分離機は1000×gで5分間、アネキシンV-FITC結合溶液中のペレットを再懸濁します。</li>

	<li>各サンプルチューブの統計を表示し、各サンプルのアポトーシス率を計算します。</li>
</ol>6. ミトコンドリア脱分極評価
<ol><li>ステップ 4.2-4.4 に従ってトリプシン化と遠心分離を行います。</li>
	<li>完全な培地の500 μLで細胞を再懸濁し、1×105 - 6 ×106 細胞に調整した細胞密度を調整します</li>
	<li>各チューブに0.5 mLのJC-1作業溶液を追加します。</li>
	<li>細胞インキュベーター中の細胞を37°Cで20分間インキュベートします。 注:インキュベーション期間中、5×JC-1染色バッファーを4mLの蒸留水に1mL添加し、JC-1染色バッファーを調製します。</li>
	<li>インキュベーション後、4°Cで3分間600×gで細胞を遠心分離する。</li>
	<li>上清を捨て、JC-1染色バッファーの1 mLにペレットを再懸濁します。</li>
	<li>手順 6.5 ~ 6.6 を 3 回繰り返します。</li>
	<li>1.5 mL遠心分離管内の氷冷染色バッファーの1 mLでHCMを再懸濁し、30分以内に分析するために細胞を使用します。</li>
	<li>PE(590 nm)検出チャネルとFITC(530 nm)を選択して、細胞内のJC-1色素の蛍光強度を測定します。 メモ:手順5.10~5.17に従ってフローサイトメーターを設定します。</li>
</ol>7. 活性酸素種アッセイ
<ol><li>ステップ 4.2-4.4 に従ってトリプシン化と遠心分離を行います。</li>
	<li>10 μM DCFDA を用いて培地中の細胞を染色し、細胞密度を 1 × 106 ~ 1 × 107 細胞に調整する</li>
	<li>37°Cで30分間インキュベートする。</li>
	<li>3~5分ごとに細胞を上下に軽くピペットします。</li>
	<li>インキュベーション後、細胞を無血清細胞培養培地で3回洗浄する。</li>
	<li>フローサイトメーターの細胞を、ステップ 5.10~5.17 に従って分析します。 注: DCF は 488 nm で励起され、530 nm で測定された発光強度です。</li>
</ol>8. カスパーゼ3/カスパーゼ8活性測定
<ol><li>ステップ 4.2 から 4.4 に従ってトリプシンを実行します。</li>
	<li>4°Cで600×gで5分間遠心分離して細胞を回収する。</li>
	<li>2個の×106細胞あたり100μLのライセートバッファーを加えます。</li>
	<li>氷の上で15分間細胞をライスします。</li>
	<li>氷浴で15分間インキュベートした後、1.6×104 x gでサンプルを遠心分離し、4°Cで15分間遠心します。</li>
	<li>上清を氷冷遠心管に移して使用します。 注:サンプル中のタンパク質濃度は、少なくとも1〜3mg/mLでなければなりません。</li>
	<li>Ac-DEVD-pNA(2 mM)を取り出し、氷浴に置いて使用してください。</li>
	<li>酵素標識プレートに40 μLのバッファー溶液を正確に添加し、サンプルを80 μL添加し、最後に10 μLのAc-DEVD-pNA(2 mM)を加えます。</li>
	<li>37°Cで120分間インキュベートします。</li>

生体内環境とインビトロ環境の間の低体温動態の違いにより、心筋細胞モデルの低体温誘導の最適なアプローチとメカニズムは不明のままである。過去数十年で、いくつかのin vitroモデルは、低温で心筋細胞を研究するために開発されました。例えば、Jana Krechらは、虚血再灌流13後の心筋アポトーシスに対する温度の影響を研究するために適度な低体温細胞モデルを確立した。多くの研究は、冷却の生理的影響に焦点を当ててきたが、再温暖化中に発生する有害な副作用の多数もあります27.これまでの研究の結果、再温暖化は、孤立した心筋細胞27,28において収縮機能不全を誘発し得る。したがって、再温暖化の温度と速度は特に不可欠です。温度の影響を厳密に制御するために、我々は、心臓手術中に使用される標準時間温度プロトコルを適用しました。このモデルでは、温度は3つの段階を含む必要な温度範囲内で正確に制御されます:1)冷却期間(1時間)。2)温度維持期間(10時間);3)温度再温暖化期間(2時間)。また、この研究で使用される温度は、穏やかな(34°C)、中程度(31°C)、および重度の(17°C)低い温度の典型的な、以前の出版物29、30、31で使用されるものと同等である。最後に、異なる温度条件が体外での心筋細胞アポトーシスに及ぼす影響に関する仮説もテストしました。予想通り、結果は、温度が有意にROSレベルを低下させ、MMPを回復し、カスパーゼ-8/カスパーゼ-3活性を低下させたことを示した。

心筋I/Rは、細胞アポプシス11の増加を伴うことはよく知られている。最近の報告では、プログラムされた心筋細胞死が開胸手術中に増加し、壊死と一致し、それによって死んだ心筋細胞の数が12に増加することが観察されている。従って、心筋細胞アポトーシスの低減は、臨床現場で有用な治療アプローチである。マウス心房HL-1心筋細胞モデルにおいて、治療下垂体は、再灌流中のチトクロムcおよびアポトーシス誘導因子(AIF)のミトコンドリア放出を減少させることが示された13。しかし、アポトーシスの調節における温度の影響は議論の余地があり、低体温症の程度に依存しているように見える。Cooperたちの研究グループは、ノルマンサーミック心肺バイパスコントロール群と比較して、深低体温循環停止を有する豚由来の心筋組織のアポトーシス率が14増加したことを観察した。さらに、いくつかの研究の結果は、深い低体温がアポトーシス経路を活性化する可能性があることを示唆しているが、あまり積極的な低体温は経路12、15、16を阻害するように見える。この結果の理由は、虚血性損傷に関連する交核化効果と、温度が心筋組織に影響を与えるメカニズムの理解の欠如に起因する可能性があります。したがって、アポトーシスが増強または減衰する温度制限は正確に定義されるべきである。

HCMの生存率に対するOGD暴露の効果は、CCK-8アッセイによって決定された。対照群で観察されたものと比較して、細胞生存率は時間依存的に有意に低下した(図2A)。再灌流後の異なる時期におけるHCMのアポトーシス率は、0から16時間までの特定の傾向を示し、アポトーシス率は徐々に増加し、16時間時点で最大速度に達した(図2B)。OGDとして12hの細胞活性を〜50%減少させたように、12hOGDは、その後の実験で細胞損傷を誘発するために使用された。
その後、アポトーシスの過程に対する温度の影響について検討した。OGD群で観察されたものと比較して、低体温症で治療した3群において生存細胞の割合が高かった(図3B)。さらに、深低体温群の細胞は、OGD群で観察された細胞よりも2.1倍高い生存率(>92%)が最も高かった。さらに、フローサイトメトリーの結果は、低体温がOGD条件下でHCMのアポトーシスを妨げていることを示した(図3A&3C)。ミトコンドリア機能障害はアポトーシスに関連しているため、ミトコンドリア障害を評価するための追加データを得た。細胞内ROSレベルはDCFH-DAアッセイを用いて決定した。OGD群で観察されたものと比較して、低体温療法はHCMにおける細胞内ROSレベルを低下させた(図4A)。また、ミトコンドリア膜電位はJC-1染色で検出された。OGD治療後、JC-1の赤色蛍光が大幅に低下し、緑色蛍光が有意に増加した。これに対し、低体温症治療はOGD誘導効果を有意に阻害し、赤対緑色比を大きなマージンで増加させた(図4B)。また、低体温治療で処理した細胞においてもカスパーゼ3/カスパーゼ-8の減少が認められた(図4C,4D)

Stem Cell Transplantation in an in vitro Simulated Ischemia/Reperfusion Model

Stem cell transplantation protocols are finding their way into clinical practice1,2,3. Getting better results, making the protocols more robust, and finding new sources for implantable cells are the focus of recent research4,5. Investigating the effectiveness of cell therapies is not an easy task and new tools are needed to investigate the mechanisms involved in the treatment process6. We designed an experimental protocol of ischemia/reperfusion in order to allow the observation of cellular connections and even subcellular mechanisms during ischemia/reperfusion injury and after stem cell transplantation and to evaluate the efficacy of cell therapy. H9c2 cardiomyoblast cells were placed onto cell culture plates7,8. Ischemia was simulated with 150 minutes in a glucose free medium with oxygen level below 0.5%. Then, normal media and oxygen levels were reintroduced to simulate reperfusion. After oxygen glucose deprivation, the damaged cells were treated with transplantation of labeled human bone marrow derived mesenchymal stem cells by adding them to the culture. Mesenchymal stem cells are preferred in clinical trials because they are easily accessible with minimal invasive surgery, easily expandable and autologous. After 24 hours of co-cultivation, cells were stained with calcein and ethidium-homodimer to differentiate between live and dead cells. This setup allowed us to investigate the intercellular connections using confocal fluorescent microscopy and to quantify the survival rate of postischemic cells by flow cytometry. Confocal microscopy showed the interactions of the two cell populations such as cell fusion and formation of intercellular nanotubes. Flow cytometry analysis revealed 3 clusters of damaged cells which can be plotted on a graph and analyzed statistically. These populations can be investigated separately and conclusions can be drawn on these data on the effectiveness of the simulated therapeutical approach.

Stem Cell Transplantation in an in vitro Simulated Ischemia/Reperfusion Model

We demonstrate how to set up an in vitro ischemia/reperfusion model and how to evaluate the effect of stem cell therapy on postischemic cardiac cells.

Ultrasonic Assessment of Myocardial Microstructure

Echocardiography is a widely accessible imaging modality that is commonly used to noninvasively characterize and quantify changes in cardiac structure and function. Ultrasonic assessments of cardiac tissue can include analyses of backscatter signal intensity within a given region of interest. Previously established techniques have relied predominantly on the integrated or mean value of backscatter signal intensities, which may be susceptible to variability from aliased data from low frame rates and time delays for algorithms based on cyclic variation. Herein, we describe an ultrasound-based imaging algorithm that extends from previous methods, can be applied to a single image frame&#160;and accounts for the full distribution of signal intensity values derived from a given myocardial sample. When applied to representative mouse and human imaging data, the algorithm distinguishes between subjects with and without exposure to chronic afterload resistance. The algorithm offers an enhanced surrogate measure of myocardial microstructure and can be performed using open-access image analysis software.

Myocardial Infarction and Functional Outcome Assessment in Pigs

Introduction of newly discovered cardiovascular therapeutics into first-in-man trials depends on a strictly regulated ethical and legal roadmap. One important prerequisite is a good understanding of all safety and efficacy aspects obtained in a large animal model that validly reflect the human scenario of myocardial infarction (MI). Pigs are widely used in this regard since their cardiac size, hemodynamics, and coronary anatomy are close to that of humans. Here, we present an effective protocol for using the porcine MI model using a closed-chest coronary balloon occlusion of the left anterior descending artery (LAD), followed by reperfusion. This approach is based on 90 min of myocardial ischemia, inducing large left ventricle infarction of the anterior, septal and inferoseptal walls. Furthermore, we present protocols for various measures of outcome that provide a wide range of information on the heart, such as cardiac systolic and diastolic function, hemodynamics, coronary flow velocity, microvascular resistance, and infarct size. This protocol can be easily tailored to meet study specific requirements for the validation of novel cardioregenerative biologics at different stages (i.e. directly after the acute ischemic insult, in the subacute setting or even in the chronic MI once scar formation has been completed). This model therefore provides a useful translational tool to study MI, subsequent adverse remodeling, and the potential of novel cardioregenerative agents.

Excitotoxic Stimulation of Brain Microslices as an In vitro Model of Stroke

Examining molecular mechanisms involved in neuropathological conditions, such as ischemic stroke, can be difficult when using whole animal systems. As such, primary or &#39;neuronal-like&#39; cell culture systems are commonly utilized. While&#160;these systems are relatively easy to work with, and are useful model systems in which various functional outcomes (such as cell death) can be readily quantified, the examined outcomes and pathways in cultured immature neurons (such as excitotoxicity-mediated cell death pathways) are not necessarily the same as those observed in mature brain, or in intact tissue. Therefore, there is the need to develop models in which cellular mechanisms in mature neural tissue can be examined. We have developed an in vitro technique that can be used to investigate a variety of molecular pathways in intact nervous tissue. The technique described herein utilizes rat cortical tissue, but this technique can be adapted to use tissue from a variety of species (such as mouse, rabbit, guinea pig, and chicken) or brain regions (for example, hippocampus, striatum, etc.). Additionally, a variety of stimulations/treatments can be used (for example, excitotoxic, administration of inhibitors, etc.). In conclusion, the brain slice model described herein can be used to examine a variety of molecular mechanisms involved in excitotoxicity-mediated brain injury.

Primary Outcome Assessment in a Pig Model of Acute Myocardial Infarction

Mortality after acute myocardial infarction remains substantial and is associated with significant morbidity, like heart failure. Novel therapeutics are therefore required to confine cardiac damage, promote survival and reduce the disease burden of heart failure. Large animal experiments are an essential part in the translational process from experimental to clinical therapies. To optimize clinical translation, robust and representative outcome measures are mandatory. The present manuscript aims to address this need by describing the assessment of three clinically relevant outcome modalities in a pig acute myocardial infarction (AMI) model: infarct size in relation to area at risk (IS/AAR) staining, 3-dimensional transesophageal echocardiography (TEE) and admittance-based pressure-volume (PV) loops. Infarct size is the main determinant driving the transition from AMI to heart failure and can be quantified by IS/AAR staining. Echocardiography is a reliable and robust tool in the assessment of global and regional cardiac function in clinical cardiology. Here, a method for three-dimensional transesophageal echocardiography (3D-TEE) in pigs is provided. Extensive insight into cardiac performance can be obtained by admittance-based pressure-volume (PV) loops, including intrinsic parameters of myocardial function that are pre- and afterload independent. Combined with a clinically feasible experimental study protocol, these outcome measures provide researchers with essential information to determine whether novel therapeutic strategies could yield promising targets for future testing in clinical studies.

Reliable and accurate outcome assessment is the key for translation of preclinical therapies into clinical treatment. The current paper describes how to assess three clinically relevant primary outcome parameters of cardiac performance and damage in a pig acute myocardial infarction model.

Myocardial Infarction in Neonatal Mice, A Model of Cardiac Regeneration

Myocardial infarction induced by coronary artery ligation has been used in many animal models as a tool to study the mechanisms of cardiac repair and regeneration, and to define new targets for therapeutics. For decades, models of complete heart regeneration existed in amphibians and fish, but a mammalian counterpart was not available. The recent discovery of a postnatal window during which mice possess regenerative capabilities has led to the establishment of a mammalian model of cardiac regeneration. A surgical model of mammalian cardiac regeneration in the neonatal mouse is presented herein. Briefly, postnatal day 1 (P1) mice are anesthetized by isoflurane and placed on an ice pad to induce hypothermia. After the chest is opened, and the left anterior descending coronary artery (LAD) is visualized, a suture is placed around the LAD to inflict myocardial ischemia in the left ventricle. The surgical procedure takes 10-15 min. Visualizing the coronary artery is crucial for accurate suture placement and reproducibility. Myocardial infarction and cardiac dysfunction are confirmed by triphenyl-tetrazolium chloride (TTC) staining and echocardiography, respectively. Complete regeneration 21 days post myocardial infarction is verified by histology. This protocol can be used to as a tool to elucidate mechanisms of mammalian cardiac regeneration after myocardial infarction.

A Flow Cytometry-based Assay for Measuring Mitochondrial Membrane Potential in Cardiac Myocytes After Hypoxia/Reoxygenation

Timely and efficient reperfusion of the occluded coronary artery is the best strategy for decreasing myocardial infarct size in patients with a ST-segment elevated myocardial infarction. However, reperfusion per se can result in further cardiomyocyte death, a phenomenon known as reperfusion injury. The opening of the mitochondrial permeability transition pore (mPTP), with the decrease of the mitochondrial membrane potential (MMP), or mitochondrial depolarization, is universally recognized as the final step of reperfusion injury and is responsible for mitochondrial and cardiomyocyte death. JC-1 is a lipophilic cationic dye that accumulates in mitochondria depending on the value of MMP. The higher the MMP is, the more JC-1 accumulates in the mitochondria. The increasing amounts of JC-1 in mitochondria can be reflected by a fluorescence emission shift from green (~530 nm) to red (~590 nm). Therefore, the reduction of the red/green fluorescence intensity ratio can indicate the depolarization of mitochondria. Here, we take advantage of JC-1 to measure MMP, or the opening of mPTP in human cardiac myocytes after hypoxia/reoxygenation, detected by flow cytometry.

Due to the limited regeneration capacity of the heart in adult mammals, myocardial infarction results in an irreversible loss of cardiomyocytes. This loss of relevant amounts of heart muscle mass can lead to the heart failure. Besides heart transplantation, there is no curative treatment option for the end-stage heart failure. In times of organ donor shortage, organ independent treatment modalities are needed. Left-ventricular assist devices are a promising therapy option, however, especially as destination therapy, limited by its side-effects like stroke, infections and bleedings. In recent years, several cardiac repair strategies including stem cell injection, cardiac progenitors or myocardial tissue engineering have been investigated. Recent improvements in cell biology allow for the differentiation of large amounts of cardiomyocytes derived from human induced pluripotent stem cells (iPSC). One of the cardiac repair strategies currently under evaluation is to transplant artificial heart tissue. Engineered heart tissue (EHT) is a three-dimensional in vitro created cardiomyocyte network, with functional properties of native heart tissue. We have created EHT-patches from hiPSC derived cardiomyocytes. Here we present a protocol for the induction of left ventricular myocardial cryoinjury in a guinea pig, followed by implantation of hiPSC derived EHT on the left ventricular wall.

Isolation of Atrial Myocytes from Adult Mice

The electrophysiological properties of atrial myocytes importantly affect overall cardiac function. Alterations in the underlying ionic currents responsible for the action potential can cause pro-arrhythmic substrates that underlie arrhythmias, such as atrial fibrillation, which are highly prevalent in many conditions and disease states. Isolating adult mouse atrial cardiomyocytes for use in patch-clamp experiments has greatly advanced our knowledge and understanding of the cellular electrophysiology in the healthy atrial myocardium and in the setting of atrial pathophysiology. In addition, studies using genetic mouse models have elucidated the role of a vast array of proteins in regulating atrial electrophysiology. Here we provide a detailed protocol for the isolation of cardiomyocytes from the atrial appendages of adult mice using a combination of enzymatic digestion and mechanical dissociation of these tissues. This approach consistently and reliably yields isolated atrial cardiomyocytes that can then be used to characterize cellular electrophysiology by measuring action potentials and ionic currents in patch-clamp experiments under a number of experimental conditions.

In Vitro Assessment of Cardiac Function Using Skinned Cardiomyocytes

In this article, we describe the steps required to isolate a single permeabilized (&#34;skinned&#34;) cardiomyocyte and attach it to a force-measuring apparatus and a motor to perform functional studies. These studies will allow measurement of cardiomyocyte stiffness (passive force) and its activation with different calcium (Ca2+)-containing solutions to determine, amongst others: maximum force development, myofilament Ca2+-sensitivity (pCa50), cooperativity (nHill) and the rate of force redevelopment (ktr). This method also enables determination of the effects of drugs acting directly on myofilaments and of the expression of exogenous recombinant proteins on both active and passive properties of cardiomyocytes. Clinically, skinned cardiomyocyte studies highlight the pathophysiology of many myocardial diseases and allow in vitro assessment of the impact of therapeutic interventions targeting the myofilaments. Altogether, this technique enables the clarification of cardiac pathophysiology by investigating correlations between in vitro and in vivo parameters in animal models and human tissue obtained during open heart or transplant surgery.
