1. 細胞培養培地(CCM)の調製

</ol>2. カル3細胞の培養
注:カル3細胞は、37°Cおよび5%CO2でT75またはT175細胞培養フラスコで培養される。細胞は7日ごとに60~80%のコンフルエンシーで、CCMは2~3日ごとに更新されます。CCMを注ぎ、新鮮なCCM(T25= 5 mL、T75 = 15 mL、およびT175 = 25 mL)をフラスコにパイプして、フラスコをインキュベーターに戻します。細胞は解凍後、実験で使用する前、または凍結する前に少なくとも2倍の経過をとるべきであり、合計で25倍以下で通過すべきである。
<ol><li>フラスコが60~80%コンフルエントであるかどうかを確認し、光顕微鏡で確認します。</li>
	<li>フラスコからCCMを注ぎます。</li>
	<li>カルシウムを含まずマグネシウムを使用せずに、5 mLの1xハンクスバランスソルト溶液(HBSS)で細胞2倍を洗浄します。洗浄を行うごとにHBSSを廃棄します。HBSSは、トリプシンを阻害する血清を除去する。</li>
	<li>T75用にトリプシン-EDTAを3 mL(T175の場合は4 mL)、フラスコを37°Cのインキュベーターに戻し、10〜15分間5%CO2 をインキュベーターに戻します。10分後に細胞がフラスコ表面から剥離していることを確認してください。細胞が>80%合流に成長した場合、トリプシン0.05%を使用してデタッチせず、トリプシン0.25%を使用することができます。</li>
	<li>フラスコにCCMの6mL(すなわち、トリプシン-EDTAボリュームを2倍)フラスコに加え、フラスコを穏やかに揺らし、適切な混合を確実にします。これは、CCMのFBSによってトリプシンが中和され、細胞上での活性が停止していることを確認するためである。トリプシンが細胞とあまりにも長く接触し続けることを許されるならば、彼らは新しい細胞培養フラスコに入れたときに再び付着しない。</li>

	<li>ヘモサイトメーターを使用して、CCMの1 mLで、死んで生きている細胞を数えます。適切なカウントのために必要な場合は、3または4 mLで細胞を希釈します。</li>
	<li>必要なCCMボリュームにセルをサスペンドし、各フラスコに細胞懸濁液を加えます。1週間で約80%の合流率を達成するために、T75に2 x 106 細胞、T175中に6 x 106 細胞をシードします。</li>

	<li>細胞が60~80%の合流率に達したら、2~3日ごとに新鮮なCCMとサブカルチャーを交換してください。</li>
</ol>3. カル3細胞を培養インサートにシーミングする
メモ:挿入は、異なる細孔サイズで利用可能です。小さな細孔サイズ(例えば、0.4 μm)は、細胞がより容易に成長し、トランス上皮電気抵抗(TEER)によって測定されるように、水没条件下で培養した5日後にすでに良好なバリアを達成できるという利点を有する。しかし、粒子転位に興味がある場合、これらの細孔は小さすぎて粒子をトラップします。したがって、大きな孔サイズ(例えば、3μm)は、通常、粒子をテストするために選択されます。より大きな孔サイズを使用する場合、細胞は良好なTEERを達成するためにより長い期間(例えば、水没条件下で培養する7〜10日)を必要とします。
<ol><li>ステップ2.1-2.10に続く既知の濃度の細胞懸濁液を調製する。</li>
	<li>細胞を5 x 105 細胞/mLの濃度に希釈し、6ウェルインサートのプレウォームCCMで、または12ウェルインサートの場合は2.5 x 105 細胞/mLの濃縮を行います。</li>

	<li>細胞培養プレートを覆い、37°Cおよび5%CO2でインキュベートする。</li>
	<li>CCM を 2 ~ 3 日ごとに変更します。</li>
	<li>アリで培養を続ける前に、細胞を水没条件下で7日間コンフルエントにします。</li>
	<li>TEER を測定します。<ol>
<li>箸電極セットを補った上皮ボルトメーターを取り、バッテリーシステムを一晩充電してください。</li>
		<li>ボルトメーターを充電器から取り外し、箸電極を接続します。</li>
		<li>70%エタノールで電極を洗浄してください。</li>
		<li>長い電極を皿の底に触れるまで外部培養培地に入れ、より短い電極を膜に触れずに媒体に入れて、CCMに電極を入れる。</li>
		<li>セルを挿入せずに空の挿入から始めます。測定が安定するまで待ち、オームズで抵抗を書き留めます。この測定は、細胞を含まない挿入膜の抵抗(すなわち、ブランク抵抗)である。</li>
		<li>各挿入物の測定を繰り返し、真の抵抗を得るためにブランク抵抗を減算します。</li>
		<li>データ解析では、抵抗値に挿入物の表面積を掛けて x cm2Ω。6ウェルインサートの場合、表面積は4.67 cm2です。したがって、600オームの抵抗が測定され、背景が120オームである場合、抵抗は480オームであり、その後、合計2,241.6オームxcm2の表面積4.67cm2を掛けた。続行するには、TEER は 1,000 Ω x cm2である必要があります。</li>

	<li>挿入部の補助側から CCM を取り外します。</li>
	<li>ウェルのバソラテラ側側に12ウェルインサート(すなわち、細胞培養インサート)に12ウェル挿入のための6ウェルのための2mLの前温化CCMの2 mLを加える。CCMは下部から膜に触れる必要がありますが、挿入物の上部に漏れ出しません。</li>
	<li>この時点で細胞は空気に対して格言的に露出し、これはALIでの培養と呼ばれる。</li>
	<li>培養器中のALIで培養細胞を37°Cで、5%CO2を曝露前7日間培養した。</li>
	<li>2~3日ごとに、バソアハラCCMを変更します。この細胞は、6週間のALIで使用することができる。</li>
</ol>4. 露出設定の準備
注: セクション 5 ~ 7 では、自動露出ステーションを使用したパーティクル露出の準備について説明します(AES、 材料表を参照)。粒子のネビュライゼーションと特性評価のセットアップは、他のメーカーの他のALI露光システムとも互換性があります。一例として、粒子への曝露を以下に説明する。このようなシステムは、感作剤、タバコの煙、ディーゼル排気などの他の露出にも使用できます。 図 1 は、AES と露出モジュールを示しています。 図2 は、他のすべての機器を含む露出設定の概略表現を示しています。
<ol><li>AESを使用して露出を開始する前に、エアロゾルの特性を測定するためにシステムをいくつかの機器に接続してください。これらは、エアロゾルがキャビネットに入る直前のサイドストリームで測定されます。 注: フロー スプリッターは、サイド ストリームを露出特性化装置に接続するために使用されます。一般的に、以下の装置が使用される:走査移動度粒子サイザー(SMPS)、光粒子サイザー(OPS)、凝縮粒子カウンター(CPC)、テーパー要素振動マイクロバランス(TEOM)。SMPSおよびOPS測定は1時間あたり1倍に実行され、フロースプリッターから同じチューブを使用します。CPCとTEOMは連続的な測定を行い、両方からのデータはSquirrelモデル2020データロガーに記録される。さらに、重量測定質量濃度は、制御された相対湿度(40~70%)のマイクロバランスを使用して決定されます。温度(21~23°C)の条件。テフロンフィルターは、各曝露の前後に計量され、曝露濃度を確認する。排気をキャプチャするために、HEPAフィルタを使用します。設計されたナノ材料(ENM)懸濁液およびネブライザーを含むセットアップはすべてフローキャビネットに置かれ、人への暴露を防ぎます。AESは金属および金属酸化物を含む多くの異なるタイプのEnmをテストするために使用することができる。</li>
	<li>露出の直前にナノ材料懸濁液を準備します。通常、1%懸濁液は、ストック溶液として調製される。例えば、100mgのナノ材料を純水10mLに懸濁させる。 注意:DQ12露光の場合、300mgを使用して約2μg/cm2を達成します。この量は、単一の暴露で使用するか、または繰り返しの暴露で分割することができます(例えば、300mgは、単一の暴露のために30mLに懸濁されるか、または21.5mgが3週間の繰り返し暴露のために毎日2.15 mLに吊り下げされます)。懸濁液は、各暴露日に新たに調製されます。粒子懸濁液は、プローブ超音波処理を使用して16分間超音波処理されます。1%ストック溶液の体積は純水を加えることによって100 mLの総容積に調節される。</li>
	<li>ENM懸濁液をキャップと磁気スターラー付きの小さなボトルに入れ、粒子の沈降を防ぎます。小さい管を介して蠕動ポンプにボトルを接続し、25 mL/hに流れを調節します。</li>
	<li>蠕動ポンプをスプレーノズルに接続し、設定を調整して連続エアロゾル化を可能にします。</li>
	<li>スプレーノズルは、長さ60cmのアルミニウムチューブ(混合チャンバー、直径15cm、60°Cに加熱)に取り付けられています。設定は1.5メートル長い銅管(直径15 cm)を介してAESに接続される。AESの上にインタクタは2.5 μmより大きいすべてのエアロゾルを取除く。</li>
	<li>スプレーノズルを2つのマスフローコントローラ(MFC)を介して3バー圧縮空気に接続し、サスペンションのネビュライゼーションを可能にします。14 L/minの1つの流れは、スプレーノズルに使用され、もう一方のMFCは、チューブ内の空気の混合に使用されます。</li>
	<li>露光開始の前日に、AESをオンにして、キャビネットが37°Cの温度に達するようにします。</li>
	<li>露光開始の2時間前にキャビネット内の空気の流れと湿度をオンにして、湿度85%に達します。セルと挿入物が配置される露出チャンバーの加熱をオンにします。</li>
	<li>石英マイクロバランス (QCM) をオンにし、ソフトウェアを使用して初期値を 0 に設定します。ログを開始します。質量は10sごとに測定され、ng/cm2として表されます。</li>
	<li>水浴で細胞培養培地を37°Cに温めます(〜20~30分)。</li>
</ol>5. 光3細胞の露出の準備
注: AES を使用した一般的な ALI エクスポージャーでは、コンフルエント セル レイヤを使用する 15 ~ 20 個の挿入が必要です。これらは、3つのクリーンエアコントロール、AESで露出せずに他のインサートと同様に処理される3つのインキュベーターコントロール、エアロゾル暴露用の6-8インサート(0、1、または2マイクロバランスの使用に応じて)、制御測定用の1-3挿入物(LDHリリースの最大放出など)、および3つの予備挿入物で構成されています。セルの TEER は 、>1,000×x cm2 Ωを持つ必要があります。
<ol><li>露光初日に、CCMで細胞1xを洗浄し、細胞の形態をチェックし、Voltohmmeterを用いて細胞モデルのTEERを測定する。細胞は、ギャップのないタイトな単層を形成する必要があります。</li>
	<li>FCSを含まない2mL/1mLのHEPES緩衝CCMを6ウェル/12ウェルプレートのバソラテラサイド側に入れ、培養インサートを新しいプレートに細胞と共に移します。 注: 露出中は、Co2 は AES に存在しません。そのため、輸送および曝露時にHEPES緩衝培養培地(25mM HEPES)が用いられる。この培地は、AES内の露出細胞およびインキュベーターコントロール細胞の両方に使用される。</li>
	<li>細胞培養物をAESに輸送する時間が5分以上である場合には、輸送中に細胞を37°Cのポータブルインキュベーターに入れる。</li>
</ol>6. 暴露時のAESの取り扱い
<ol><li>AESでは、胎児子牛血清(FCS)なしでHEPES緩衝CCMで露光モジュールを充填します。CCMの量は、使用される単位と細胞培養挿入物の種類によって異なります。アリで細胞を維持するために、媒体は膜の底に達するべきであるが、膜の上に漏れてはならないことを覚えておいてください。6つの井戸挿入物を使用する場合、6 mLのHEPESバッファリングCCMを露光モジュールに加えます。</li>
	<li>無菌ピンセットを使用して、プレートから露出モジュールにセルを持つ挿入物を転送します。細胞のバソラテラ側面に気泡がないことを確認し、挿入物の補助側のCCMを取り除きます。バソラテラ側に気泡がある場合は、滅菌ピンセットを使用して挿入物をそっと回します。CCMを含むプレートを、露光後の転写用インキュベーターに保管してください。</li>
	<li>タッチスクリーンディスプレイを使用して、露出時間、空気流量、静電堆積の向上を選択します。また、ディスプレイは湿度や温度を確認するために使用することができます。通常、37 °Cおよび85%の湿度で挿入物の流量50 mL/minで、4時間の露光時間が選択されます。 注: 第 1 レベルのモジュール (図 1) は、クリーンエア露出に使用されます。このレベルのインサートは、クリーンエア露出制御として使用されます。第2および第3レベルの他のモジュールは、水晶結晶マイクロバランス(QCM)がオンラインで堆積を測定するための2つのモジュールを含むエアロゾル暴露に使用することができます。</li>
	<li>漏れ検査は、露光を開始する前に行う必要があります。漏出は5 mL/min未満である必要があります。AES ディスプレイの指示に従います。リークテストが終了したら、露光を開始できます。漏れの場合は、チューブをチェックする必要があります。</li>
	<li>露光の終わりに、AESモジュールのドアを開け、露光モジュールを開き、細胞培養挿入物を細胞培養プレートに戻し、プレートをポータブルインキュベーターに移します。</li>
	<li>モジュール(すなわち、露出したサンプル)およびプレート(すなわち、インキュベーター制御)から、乳酸脱水素酵素(LDH)測定など後で分析するために、培地を収集する。</li>
	<li>細胞培養ラボに戻り、新鮮な標準CCMを充填したプレートに細胞培養インサートを移します。細胞培養プレートを次の暴露まで、または分析するまでインキュベーターに入れます。</li>
	<li>最終露光日の後、インキュベーターにインサートを翌日まで入れます。</li>
	<li>最終露出の翌日に、CCMを補助側に加えて、Voltohmmeterを使用してTEERを測定します。サイトカインの分析のために、補助的なCCMとバソラショナルCCMの両方を別々に収集します。</li>
	<li>すべてのCCMを除去し、例えば、アプリカル側に増殖試薬を加えることによって細胞生存アッセイを行う。</li>
</ol>7. AES のクリーニング

次に、物質に対するCalu-3モデルの応答を試験した。陽性対照物質として、LPSは、モデルの尖側にネビュライゼーションを介して投与した。沈着した用量は0.25 μg/cm2であった。カル3細胞はLDH放出の増加と腫瘍壊死因子α(TNF-α)放出によるリポ多糖(LPS)への反応を示した(図6)。
最後に、カル3単層を石英シリカ(DQ12)ナノ材料(IOM、エディンバラ)にさらした。結晶性シリカは珪肺を誘発し、肺腫瘍を引き起こす可能性もある。そこで、国際がん研究機関(IARC)は、結晶性シリカをグループIヒト発がん性物質20に分類した。結晶性シリカの作用機序は、その反応性表面21、22、23によって引き起こされる持続性炎症の誘導を介してであると考えられる。ラットおよびマウスの両方におけるいくつかのインビボ研究は、結晶性シリカ24、25、26、27、28、29への吸入暴露後に、腫瘍および線維症を含む炎症および組織病理学的変化の誘導を報告する。これらの効果はすべて、繰り返し暴露および/または長期フォローアップ後に観察される。Calu-3モデルは、ALIで繰り返し暴露できるインビトロモデルを使用して、生体内研究からの観測を模倣できるかどうかを調べるのに使用されました。
カル3細胞を3週連続で、週5日、DQ12に1日あたり4時間曝露した。堆積した用量はQCMを用いて測定した。平均沈着用量は1日あたり120 ng/cm2 で、累積用量は1.6 μg/cm2で、インビボで効果を誘発する用量と同様であった。その他の粒子特性を 表1に示す。3週間の曝露後、DQ12は、清浄空気制御と比較して、TEER、細胞生存率、及び単球化学誘引タンパク質1(MCP-1)放出において有意な効果を誘発しなかった(図7)。インビボデータに基づいてDQ12の毒性が予想されるほど、EUプロジェクトGRACIOUS(成果物5.3)内で最適化されたプロトコルに従って、粒子の反応性を無細胞アッセイを用いてチェックした。DQ12バッチの反応性は予想よりも低く(図8)、正の対照粒子カーボンブラック(CB)に比べて桁違い低かった。この反応性の欠如は、Calu-3モデルにおける毒性応答の欠如を説明するかもしれない。

Pulmonary epithelial cells can be isolated from the respiratory tract of mice and cultured at air-liquid interface (ALI) as a model of differentiated respiratory epithelium. A protocol is described for isolating and exposing these cells to mainstream cigarette smoke (CS), in order to study epithelial cell responses to CS exposure. The protocol consists of three parts: the isolation of airway epithelial cells from mouse trachea, the culturing of these cells at air-liquid interface (ALI) as fully differentiated epithelial cells, and the delivery of calibrated mainstream CS to these cells in culture. The ALI culture system allows the culture of respiratory epithelia under conditions that more closely resemble their physiological setting than ordinary liquid culture systems. The study of molecular and lung cellular responses to CS exposure is a critical component of understanding the impact of environmental air pollution on human health. Research findings in this area may ultimately contribute towards understanding the etiology of chronic obstructive pulmonary disease (COPD), and other tobacco-related diseases, which represent major global health problems.

A New Portable In Vitro Exposure Cassette for Aerosol Sampling

This protocol introduces a new in vitro exposure system, capable of being worn, including its characterization and performance. Air-liquid interface (ALI) in vitro exposure systems are often large and bulky, making transport to the field and operation at the source of emission or within the breathing zone difficult. Through miniaturization of these systems, the lab can be brought to the field, expediting processing time and providing a more appropriate exposure method that does not alter the aerosol prior to contacting the cells. The Portable In vitro Exposure Cassette (PIVEC) adapts a 37 mm filter cassette to allow for in vitro toxicity testing outside of a traditional laboratory setting. The PIVEC was characterized using three sizes of copper nanoparticles to determine deposition efficiency based on gravimetric and particle number concentration analysis. Initial cytotoxicity experiments were performed with exposed lung cells to determine the ability of the system to deposit particles while maintaining cell viability. The PIVEC provides a similar or increased deposition efficiency when comparing to available perpendicular flow in vitro exposure devices. Despite the lower sample throughput, the small size gives some advantages to the current in vitro ALI exposure systems. These include the ability to be worn for personal monitoring, mobility from the laboratory to the source of emission, and the option to set-up multiple systems for spatial resolution while maintaining a lower user cost. The PIVEC is a system capable of collecting aerosols in the field and within the breathing zone onto an air-interfaced, in vitro model.

In vitro Cell Culture Model for Toxic Inhaled Chemical Testing

Cell cultures are indispensable to develop and study efficacy of therapeutic agents, prior to their use in animal models. We have the unique ability to model well differentiated human airway epithelium and heart muscle cells. This could be an invaluable tool to study the deleterious effects of toxic inhaled chemicals, such as chlorine, that can normally interact with the cell surfaces, and form various byproducts upon reacting with water, and limiting their effects in submerged cultures. Our model using well differentiated human airway epithelial cell cultures at air-liqiuid interface circumvents this limitation as well as provides an opportunity to evaluate critical mechanisms of toxicity of potential poisonous inhaled chemicals. We describe enhanced loss of membrane integrity, caspase release and death upon toxic inhaled chemical such as chlorine exposure. In this article, we propose methods to model chlorine exposure in mammalian heart and airway epithelial cells in culture and simple tests to evaluate its effect on these cell types.

Impact Assessment of Repeated Exposure of Organotypic 3D Bronchial and Nasal Tissue Culture Models to Whole Cigarette Smoke

Cigarette smoke (CS) has a major impact on lung biology and may result in the development of lung diseases such as chronic obstructive pulmonary disease or lung cancer. To understand the underlying mechanisms of disease development, it would be important to examine the impact of CS exposure directly on lung tissues. However, this approach is difficult to implement in epidemiological studies because lung tissue sampling is complex and invasive. Alternatively, tissue culture models can facilitate the assessment of exposure impacts on the lung tissue. Submerged 2D cell cultures, such as normal human bronchial epithelial (NHBE) cell cultures, have traditionally been used for this purpose. However, they cannot be exposed directly to smoke in a similar manner to the in vivo exposure situation. Recently developed 3D tissue culture models better reflect the in vivo situation because they can be cultured at the air-liquid interface (ALI). Their basal sides are immersed in the culture medium; whereas, their apical sides are exposed to air. Moreover, organotypic tissue cultures that contain different type of cells, better represent the physiology of the tissue in vivo. In this work, the utilization of an in vitro exposure system to expose human organotypic bronchial and nasal tissue models to mainstream CS is demonstrated. Ciliary beating frequency and the activity of cytochrome P450s (CYP) 1A1/1B1 were measured to assess functional impacts of CS on the tissues. Furthermore, to examine CS-induced alterations at the molecular level, gene expression profiles were generated from the tissues following exposure. A slight increase in CYP1A1/1B1 activity was observed in CS-exposed tissues compared with air-exposed tissues. A network-and transcriptomics-based systems biology approach was sufficiently robust to demonstrate CS-induced alterations of xenobiotic metabolism that were similar to those observed in the bronchial and nasal epithelial cells obtained from smokers.

The aim of this protocol is to expose human organotypic 3D bronchial and nasal tissue models to mainstream cigarette smoke (CS) at the air-liquid interface. The impact of CS on the tissues is then investigated using a cytochrome P450 activity assay, a cilia beating measurement, and a systems biology approach.

A Reversible, Non-invasive Method for Airway Resistance Measurements and Bronchoalveolar Lavage Fluid Sampling in Mice

Airway hyperreactivity (AHR) measurements and bronchoalveolar lavage (BAL) fluid sampling are essential to experimental asthma models, but repeated procedures to obtain such measurements in the same animal are generally not feasible.  Here, we demonstrate protocols for obtaining from mice repeated measurements of AHR and bronchoalveolar lavage fluid samples.  Mice were challenged intranasally seven times over 14 days with a potent allergen or sham treated.  Prior to the initial challenge, and within 24 hours following each intranasal challenge, the same animals were anesthetized, orally intubated and mechanically ventilated.  AHR, assessed by comparing dose response curves of respiratory system resistance (RRS) induced by increasing intravenous doses of acetylcholine (Ach) chloride between sham and allergen-challenged animals, were determined.  Afterwards, and via the same intubation, the left lung was lavaged so that differential enumeration of airway cells could be performed.  These studies reveal that repeated measurements of AHR and BAL fluid collection are possible from the same animals and that maximal airway hyperresponsiveness and airway eosinophilia are achieved within 7-10 days of initiating allergen challenge.  This novel technique significantly reduces the number of mice required for longitudinal experimentation and is applicable to diverse rodent species, disease models and airway physiology instruments.  

Repeated measurements of rodent respiratory physiology and sampling of airway inflammatory cells are desirable, but generally not feasible. Here we describe a repeatable method for orally intubating mice that permits repeated measurements of airway hyperreactivity and sampling of airway inflammatory cells.

P. aeruginosa Infected 3D Co-Culture of Bronchial Epithelial Cells and Macrophages at Air-Liquid Interface for Preclinical Evaluation of Anti-Infectives

fDrug research for the treatment of lung infections is progressing towards predictive in vitro models of high complexity. The multifaceted presence of bacteria in lung models can re-adapt epithelial arrangement, while immune cells coordinate an inflammatory response against the bacteria in the microenvironment. While in vivo models have been the choice for testing new anti-infectives in the context of cystic fibrosis, they still do not accurately mimic the in vivo conditions of such diseases in humans and the treatment outcomes. Complex in vitro models of the infected airways based on human cells (bronchial epithelial and macrophages) and relevant pathogens could bridge this gap and facilitate the translation of new anti-infectives into the clinic. For such purposes, a co-culture model of the human cystic fibrosis bronchial epithelial cell line CFBE41o- and THP-1 monocyte-derived macrophages has been established, mimicking an infection of the human bronchial mucosa by P. aeruginosa at air-liquid interface (ALI) conditions. This model is set up in seven days, and the following parameters are simultaneously assessed: epithelial barrier integrity, macrophage transmigration, bacterial survival, and inflammation. The present protocol describes a robust and reproducible system for evaluating drug efficacy and host responses that could be relevant for discovering new anti-infectives and optimizing their aerosol delivery to the lungs.

We describe a protocol for a three-dimensional co-culture model of infected airways, using CFBE41o- cells, THP-1 macrophages, and Pseudomonas aeruginosa, established at the air-liquid interface. Isolating Bronchial Epithelial Cells from Resected Lung Tissue for Biobanking and Establishing Well-Differentiated Air-Liquid Interface Cultures

The airway epithelial cell layer forms the first barrier between lung tissue and the outside environment and is thereby constantly exposed to inhaled substances, including infectious agents and air pollutants. The airway epithelial layer plays a central role in a large variety of acute and chronic lung diseases, and various treatments targeting this epithelium are administered by inhalation. Understanding the role of epithelium in pathogenesis and how it can be targeted for therapy requires robust and representative models. In vitro epithelial culture models are increasingly being used and offer the advantage of performing experiments in a controlled environment, exposing the cells to different kinds of stimuli, toxicants, or infectious agents. The use of primary cells instead of immortalized or tumor cell lines has the advantage that these cells differentiate in culture to a pseudostratified polarized epithelial cell layer with a better representation of the epithelium compared to cell lines.
Presented here is a robust protocol, that has been optimized over the past decades, for the isolation and culture of airway epithelial cells from lung tissue. This procedure allows successful isolation, expansion, culture, and mucociliary differentiation of primary bronchial epithelial cells (PBECs) by culturing at the air-liquid interface (ALI) and includes a protocol for biobanking. Furthermore, the characterization of these cultures using cell-specific marker genes is described. These ALI-PBEC cultures can be used for a range of applications, including exposure to whole cigarette smoke or inflammatory mediators, and co-culture/infection with viruses or bacteria. 
The protocol provided in this manuscript, illustrating the procedure in a step-by-step manner, is expected to provide a basis and/or reference for those interested in implementing or adapting such culture systems in their laboratory.

Presented here is a reproducible, affordable, and robust method for the isolation and expansion of primary bronchial epithelial cells for long-term biobanking and the generation of differentiated epithelial cells by culture at the air-liquid interface.

Assessment of the Acute Inhalation Toxicity of Airborne Particles by Exposing Cultivated Human Lung Cells at the Air-Liquid Interface

Here, we present a specially designed modular in vitro exposure system that enables the homogenous exposure of cultivated human lung cells at the ALI to gases, particles or complex atmospheres (e.g., cigarette smoke), thus providing realistic physiological exposure of the apical surface of the human alveolar region to air. In contrast to sequential exposure models with linear aerosol guidance, the modular design of the radial flow system meets all requirements for the continuous generation and transport of the test atmosphere to the cells, a homogenous distribution and deposition of the particles and the continuous removal of the atmosphere. This exposure method is primarily designed for the exposure of cells to airborne particles, but can be adapted to the exposure of liquid aerosols and highly toxic and aggressive gases depending on the aerosol generation method and the material of the exposure modules.
Within the framework of a recently completed validation study, this exposure system was proven as a transferable, reproducible and predictive screening method for the qualitative assessment of the acute pulmonary cytotoxicity of airborne particles, thereby potentially reducing or replacing animal experiments that would normally provide this toxicological assessment.

Establishing a Silicosis Rat Model via Exposure of Whole-Body to Respirable Silica

The major cause of silicosis is the inhalation of silica in the occupational environment. Despite some anatomical and physiological differences, rodent models continue to be an essential tool for studying human silicosis. For silicosis, the classic pathological process needs to be inducible via the inhalation of freshly generated quartz particles, which means specifically inducing human occupational disease. This study described a technique to establish and effectively mimic the pathological dynamic evolution process of silicosis. Further, the technique had good repeatability with no surgery involved. The inhalation exposure system was fabricated, validated, and used for toxicology studies on respirable particle inhalation. The critical components were as follows: (1) bulk dry SiO2 powder generator adjusted with an air-flow controller; (2) 0.3 m3 whole-body inhalation exposure chamber accommodating up to 3&#160;adult rats; (3) a monitoring and control system for regulating oxygen concentration, temperature, humidity, and pressure in real-time; and (4) a barrier and waste disposal system for protecting laboratory technicians and the environment. In summary, the present protocol reports the inhalation via the whole body, and the inhalation chamber created a reliable, reasonable, and repeatable rat silicotic model with low mortality, less injury, and more protection.

This study describes a technique to establish a silicosis rat model with the inhalation of silica through the whole body in an inhalation chamber. The rats with silicosis could closely mimic the pathological process of human silicosis in an easy, cost-effective manner with good repeatability.

Multicellular Human Alveolar Model Composed of Epithelial Cells and Primary Immune Cells for Hazard Assessment

A human alveolar cell coculture model is described here for simulation of the alveolar epithelial tissue barrier composed of alveolar epithelial type II cells and two types of immune cells (i.e., human monocyte-derived macrophages [MDMs] and dendritic cells [MDDCs]). A protocol for assembling the multicellular model is provided. Alveolar epithelial cells (A549 cell line) are grown and differentiated under submerged conditions on permeable inserts in two-chamber wells, then combined with differentiated MDMs and MDDCs. Finally, the cells are exposed to an air-liquid interface for several days. As human primary immune cells need to be isolated from human buffy coats, immune cells differentiated from either fresh or thawed monocytes are compared in order to tailor the method based on experimental needs. The three-dimensional models, composed of alveolar cells with either freshly isolated or thawed monocyte-derived immune cells, show a statistically significant increase in cytokine (interleukins 6 and 8) release upon exposure to proinflammatory stimuli (lipopolysaccharide and tumor necrosis factor &#945;) compared to untreated cells. On the other hand, there is no statistically significant difference between the cytokine release observed in the cocultures. This shows that the presented model is responsive to proinflammatory stimuli in the presence of MDMs and MDDCs differentiated from fresh or thawed peripheral blood monocytes (PBMs). Thus, it is a powerful tool for investigations of acute biological response to different substances, including aerosolized drugs or nanomaterials.

Presented here is a protocol for primary human blood monocyte isolation as well as their differentiation into macrophages and dendritic cells and assembly with epithelial cells into a multicellular human lung model. Biological responses of cocultures composed of immune cells differentiated from either freshly isolated or thawed monocytes, upon exposure to proinflammatory stimuli, are compared.
