1. Vorbereitung von Substraten
<ol><li>Verwenden Sie eine Substratoberfläche, die für die Anwendung geeignet ist. Verwenden Sie für Holz ein geschnittenes Furnier von etwa 0,6 bis 0,8 mm Dicke von einem zuverlässigen Hersteller, da diese Furniere für die Herstellung von Sperrholz und Furnierschichtholz (LVL) verwendet werden. Diese werden von einem Furnierlieferanten als Platten von 0,6 bis 0,8 mm Dicke bezogen und seitlich in 305 mm geschnitten. Ein konsistentes Substrat ist ein hartes Ahornfurnier (Acer saccharum) aufgrund seiner Oberflächenglätte und konsistenten Dicke, und es ist ein diffuses poröses Hartholz mit hohem Modul. Ahornfurniere werden häufig im Schrankbau verwendet und sind in der Regel frei von Mängeln.</li>
	<li>Konditionieren Sie das ungestapelte Holz mindestens einen Tag vor der Verwendung bei 21 °C und 50% relativer Luftfeuchtigkeit (rF). Vermeiden Sie Veneers, die übermäßig wellig sind, eine unebene Oberfläche haben und Defekte einschließlich Verfärbungen enthalten. HINWEIS: Andere Holzarten können verwendet werden, um die Haftleistung des Klebstoffs mit diesen Arten zu verstehen. Diffusporöse Harthölzer und Nadelhölzer mit einem allmählichen Übergang von Frühholz zu Spätholz werden jedoch wegen ihrer Gleichmäßigkeit empfohlen. Seien Sie vorsichtig, denn Holz kann sauer oder basisch sein oder Extrakte auf der Oberfläche enthalten, die den Klebstoffhärtungsprozess verändern können. Darüber hinaus kann die Verarbeitung des Baumes vom Zeitpunkt des Schneidens bis zur Furnierherstellung die Klebefestigkeit12,13 verändern. Da der ABES eine geringe Menge Holz verwendet, wird er weniger von Holzschwankungen beeinflusst, die bei anderen Tests auftreten, wie z. B. Holzfeuchtegehalt und Furnierprüftiefe.</li>
	<li>Stellen Sie sicher, dass die Seiten des Furniers frei von losen Fasern entlang der Kante sind und das geklebte Produkt keinen signifikanten Klebstoffdruck aufweist, da diese dazu neigen, die Klebefestigkeit zu überschätzen, da die Proben nach dem Kleben nicht modifiziert werden.</li>
</ol>2. Vorbereitung der Proben
<ol><li>Konditionieren Sie die Holzproben mindestens einen Tag lang bei 21 °C und 50 % relative Luftfeuchtigkeit. Überprüfen Sie das Furnier auf Risse, Verfärbungen oder Narbenunregelmäßigkeiten, die beim Schneiden der Proben vermieden werden sollen.</li>
	<li>Stellen Sie sicher, dass die pneumatisch angetriebene Probenschneidvorrichtung betriebsbereit ist.</li>
	<li>Verwenden Sie eine spezielle Stanze, die die erforderliche Probengröße von 20 mm x 117 mm von 0,6 auf 0,8 mm dickes Ahornfurnier schneidet (Abbildung 1, Materialtabelle).<ol><li>Legen Sie ein Stück Furnier, mindestens 150 mm x 300 mm, unter die Schneidmesser, so dass die Furniermaserung parallel zur langen Richtung verläuft, und drücken Sie den Luftdruckknopf, um jedes Stück Holz von 20 mm x 117 mm zu schneiden.</li>
		<li>Bewegen Sie das Furnierstück unter den Schneidklingen an einen ungeschnittenen Bereich und drücken Sie den Knopf erneut, um ein weiteres Stück Holz zu schneiden. Fahren Sie fort, bis das Furnierstück vollständig in Stücke geschnitten ist. HINWEIS: Wenn die lange Richtung der Probe nicht parallel zur Kornrichtung verläuft, kann während eines Versuchs ein früher Bruch im Holz außerhalb des gebundenen Teils auftreten.</li>
	</ol></li>
	<li>Bei anderen Materialien als Holz schneiden Sie die Proben mit den entsprechenden Techniken. Wenn das Material nicht mit dem Probenschneider geschnitten werden kann, verwenden Sie das Material, um es auf die erforderliche Größe zu schneiden. Aufgrund der kleinen Klebefläche ist es wichtig, dass das Schneiden präzise und die Proben frei von Ablagerungen entlang der Kanten und auf den Klebeflächen sind.</li>
</ol>3. Funktionsfähigkeit der Geräte
<ol><li>Stellen Sie für den Klebevorgang sicher, dass die ABES-Ausrüstung gemäß einer Standardarbeitsanweisung11 ordnungsgemäß funktioniert. Die Einstellungen auf der Vorderseite der ABES-Einheit zum Kleben und Brechen von Proben sind: LP Press 0,2 MPa, HP Press 0,2 MPa, Pull 0,65 MPa und Cool Air 0,2 MPa.</li>
	<li>Verwenden Sie einen Luftversorgungsdruck von mindestens 0,62 MPa (90 psig), da ein zu niedriger Druck dazu führt, dass sich die Greifklemmen und -platten zu langsam oder ungleichmäßig auf der Probe schließen, was zu falschen Verbundfestigkeiten führt (Abbildung 2, oben).</li>
	<li>Reinigen Sie die Platten von allen Klebstoffen, die durch Auspressen aus der vorherigen Probe entstehen. Stellen Sie die Temperatur der Aufspannplatten auf die gewünschte Temperatur ein und gleichen Sie sie aus, bevor Sie die Proben verkleben.</li>
	<li>Um Holz zu verkleben, betreiben Sie die Geräte in einem Raum mit 21 °C und 50% relative Luftfeuchtigkeit. Wenn dies nicht möglich ist, bewahren Sie die konditionierten Proben bis zur Verklebung in einem Plastikbeutel auf, da sich die Holzfeuchtigkeit aufgrund der geringen Größe der Proben schnell ändert.</li>
	<li>Um kinetische Aushärtungsdaten zu erhalten, ist das Verfahren so zu konstruieren, dass die mechanischen und elektronischen Geschwindigkeiten ausreichen, um Daten genau zu erfassen, wie in ASTM D7998-1911 beschrieben.</li>
</ol>4. Verklebung der Proben mit dem Klebstoff
HINWEIS: Die Anwendung des Klebstoffs ist ein kritisches Problem für Holzklebstoffe aufgrund der großen Variation der Viskosität und des prozentualen Feststoffanteils, die von einem Laminierklebstoff wie in Sperrholz zu einem sprühfähigen Klebstoff für Bindemittelanwendungen gehen. Holzklebstoffe sind in der Regel wässrig, so dass die Verdunstung nur ein geringes Problem darstellt. Es ist jedoch wichtig, dass Wasser in das poröse Holz eindringt.
<ol><li>5 mg des zu untersuchenden Klebstoffs über die Klemme 0,5 cm ausreichend verteilen, um den Klebebereich abzudecken und auf die andere Probe zu übertragen, jedoch ohne übermäßiges Auspressen. Um eine relativ konstante Klebstoffstreurate zu erhalten, ist die Holzprobe auf einer Waage zu halten und nach dem Klebstoffauftrag erneut zu wiegen.</li>
	<li>Verteilen Sie den Klebstoff mit großer Sorgfalt, überlappen Sie die Proben und stellen Sie sicher, dass die beiden Proben ausgerichtet sind, da ein kleiner Klebebereich verwendet wird und die Festigkeiten als Zugkraft über den Klebebereich bestimmt werden (Abbildung 2 unten). Es können unterschiedliche Klebebereiche verwendet werden, aber die Festigkeit ist aufgrund der unterschiedlichen Mechanik von Überrundungsscherversuchen nicht unbedingt vergleichbar. HINWEIS: Die Literatur empfiehlt mehrere Möglichkeiten, den Klebstoff je nach Klebstoffkonsistenz auf das Holz aufzutragen. Die ursprünglich empfohlene Klebstoffauftragsmethode verwendete eine speziell entwickelte Mikrospritzvorrichtung10, die sich jedoch als unordentlich, langsam und sehr abhängig von der Klebstoffrheologie erwies. Obwohl bei diesem Verfahren der Klebstoff als diskrete Punkte aufgebracht wird, wie sie in Bindemittelanwendungen für Spanplatten und orientierte Streifenplatten verwendet werden, scheint ein Druckverfahren zuverlässigerzu sein 14. Das Mikropipettenauftragsverfahren kann ein reproduzierbares Klebstoffvolumen10 liefern, aber es ist etwas schwierig, es gleichmäßig zu verteilen. Die Spachtelmethode hat sich am besten bewährt, um eine gleichmäßige Verteilung des Klebstoffs auf der Klebefläche zu erhalten, und eine Mikrowaage zur Erzielung einer gemessenen Menge wird empfohlen11.</li>
	<li>Endfestigkeitsdaten<ol><li>Die Proben bei 120 °C für 2 min verkleben und über Nacht bei 21 °C und 50% rF konditionieren, da das Holz durch das Heißpressen während des Verklebens austrocknet. Um das Holz zu verkleben, verriegeln Sie eine Probe, indem Sie die Griffe am ABES-Tester schließen und sicherstellen, dass die Probe mit dem Tester ausgerichtet ist. Drücken Sie dann den Startknopf an der Maschine, damit die 120 °C-Aufspannplatten 2 Minuten lang auf den überlappenden Abschnitt drücken, bevor Sie die Aufspannplatten zurückziehen und die Griffe lösen, damit die Proben entnommen werden können. HINWEIS: Die Zeit und Temperatur für die Aushärtung werden von der Anwendung und der Klebstoffchemie bestimmt. Die Klebetemperatur und -zeit sollte so optimiert werden, dass die Festigkeit das höchste Plateau erreicht, indem verschiedene Bindungstemperaturen und -zeiten verwendet werden, um Bedingungen für maximale Festigkeit zu bestimmen. Für Holzverbindungen ist die Prüfung der Trockenscherfestigkeit wertvoll, aber Nasstests sind im Allgemeinen kritischer, um die Haltbarkeit des Klebstoffs zu bestimmen, und erfordern ein 4-stündiges Einweichen der Probe bei Raumtemperatur in Wasser.</li>
		<li>Verriegeln Sie zum Testen eine Probe, indem Sie die Griffe am ABES-Tester schließen und sicherstellen, dass die Probe mit dem Tester ausgerichtet ist. Durch Drücken des Startknopfes zieht das Instrument an einem Ende durch einen Servoantrieb, während das andere Ende der Probe an einer an den Griffen befestigten Wägezelle zieht. Dieses Ziehen setzt sich fort, bis die Verbindung bricht. Der Computer zeichnet die maximale Kraft auf, der die Probe standhalten kann, die als Haftfestigkeit aufgezeichnet wird.<ol><li>Verwenden Sie das gleiche Verfahren für die trockenen und wassergetränkten Proben. Achten Sie bei der Messung der Bruchkraft darauf, dass die Griffe das Holz fest halten, denn wenn der Klebstoff sehr stark ist, kann das Holz verrutschen. Wenn die Probe außerhalb des gebundenen Bereichs bricht, verwerfen Sie den Wert, da dies die Holzfestigkeit und nicht den Klebstoff misst.</li>
		</ol></li>
	</ol></li>
	<li>Kinetische Kraftentwicklung<ol><li>Bestimmen Sie die Festigkeitsentwicklung eines Klebstoffs, um die für große Produkte erforderliche Presszeit abzuschätzen. Befolgen Sie das gleiche Verfahren wie in Schritt 4.3, außer dass Temperatur und Zeit geändert werden. Beginnen Sie mit der Prüfung der Festigkeit bei 100 °C Plattentemperatur mit Klebezeiten von 10, 30, 60, 90, 120, 150, 180 und 210 Sekunden. Anschließend die Temperatur um 10 °C erhöhen und die Klebezeiten wiederholen, bis bei den niedrigen Bindungszeiten kein linearer Festigkeitsquerschnitt mehr vorhanden ist.</li>
		<li>Nach dem Verkleben ziehen Sie die Platten zurück und verwenden die Luftkühlungsfunktion des ABES, um die Probe auf nahezu Raumtemperatur abzukühlen und dann die Probenstärke zu messen. Indem Sie mit einer niedrigen Presszeit beginnen und zuerst die Zeit für nachfolgende Proben erhöhen, sammeln Sie die Festigkeits-Zeit-Daten, bis eine zunehmende Zeit zu wenig oder keiner zunehmenden Festigkeit führt. Wenn Sie dann die gleiche Sequenz bei höheren Temperaturen durchführen, erhalten Sie das resultierende Diagramm der Festigkeit über die Zeit und die Aushärtungsrate als die Steigung (Abbildung 3). HINWEIS: Die phenolischen Klebstoffdaten in Abbildung 3a10 zeigen den Einfluss der Temperatur auf die Festigkeitsentwicklung zu verschiedenen Zeitpunkten. Abbildung 3b zeigt die regressierte isotherme Festigkeitsentwicklungsrate in Abhängigkeit von der Temperatur. Um die isotherme Festigkeitsentwicklung zu erhalten, wurde die Probe vor dem Test abgekühlt. Einige Klebstoffe, wie Harnstoffformaldehyd15, haben eine optimale Klebezeit und -temperatur, bevor der Abbau beginnt. Diese Methode kann dieses Problem erkennen und optimale Bedingungen bestimmen.</li>
	</ol></li>
	<li>Wärmebeständigkeit<ol><li>Wenn das Produkt eine bestimmte Temperaturbeständigkeit erfüllen muss, klemmen Sie die gebundene Probe in die ABES-Einheit. Nachdem die Platten auf diese Temperatur, z. B. 220 °C, erhitzt worden sind, oberhalb derer das Holz zu zersetzen beginnt, schließen Sie sie für 2 min auf die vorgebundene Probe und öffnen Sie sie dann, um die Klebkraft gemäß 4.3.2 zu messen, um eine etwaige thermische Erweichung des Klebstoffs im Vergleich zur Klebetemperatur von 120 °C zu bestimmen.</li>
		<li>Wiederholen Sie diesen Test, mit der Ausnahme, dass die Aufspannplatten auf der Probe für 30 Minuten geschlossen und dann auf Festigkeit getestet werden, um die Festigkeit zu bestimmen, wenn der Klebstoff thermisch abgebaut ist. Die Freigabe der Platten und die Prüffestigkeit bestimmen die Wärmebeständigkeit der Probe im Vergleich zum Wert vor dem Erhitzen. Diese Art von Verfahren wurde verwendet, um Holzklebstoffezu testen 16. Da das ABES eine schnelle Erwärmung verwendet und die Festigkeit messen kann, während es heiß ist, ohne die Probe zu einer anderen Maschine zu bewegen, kann es verwendet werden, um zwischen den beiden Fehlerarten (d. H. Thermische Erweichung oder Degradation) zu unterscheiden. Die thermische Enthärtung führt unmittelbar nach dem Erhitzen zu einem Festigkeitsverlust und ist in der Regel wiederherstellbar. Der chemische Abbau erfolgt allmählich im Laufe der Zeit bei hohen Temperaturen und gewinnt die mechanische Festigkeit beim Abkühlen nicht zurück. HINWEIS: Klebstoffhersteller müssen unterscheiden, ob der Festigkeitsverlust auf thermische Erweichung oder chemischen Abbau zurückzuführen ist, da diese Probleme unterschiedliche Lösungen erfordern. Es gibt viele Methoden, die Erweichungsübergänge messen können, einschließlich anderer thermischer Analysen, aber sie unterscheiden nicht zwischen einer Änderung der mechanischen Eigenschaften und der chemischen Struktur.</li>

</ol>5. Bildanalyse der fehlgeschlagenen Klebefläche
<ol><li>Da das Hauptziel darin besteht, die Haftfestigkeit oder die Geschwindigkeit der kohäsiven Festigkeitsentwicklung zu bestimmen, stellen Sie sicher, dass das Versagen innerhalb des Klebstoffs und nicht durch Haftung am Substrat (Abbildung 4) oder Substratversagen erfolgt. Wenn ein Substratversagen auftritt, hat der Klebstoff eine ausreichende Festigkeit. Alternativ weist ein kohäsives Versagen des Schüttklebstoffs auf eine Klebstoffschwäche hin. Die Entscheidung zwischen Adhäsion und adhäsivem Interphasenversagen kann jedoch schwierig sein17. Für die Holzanalyse wurden verschiedene Methoden entwickelt18.</li>

Kritische Schritte im Verfahren sind wie folgt: Auswahl der Substrate, Vorbereitung der Proben, Funktionsfähigkeit der Ausrüstung und Verklebung der Proben.
Das Substrat muss stark sein, minimale Defekte aufweisen (glatt, flach, keine Risse und keine Verfärbungen). Ungeschliffenes, rotierend geschnittenes Schrankfurnier aus diffusem porösem Hartholz mit Zuckerahorn (Acer saccharum) bevorzugt. Durch das Schleifen entsteht eine weniger ebene und fragmentiertere Oberfläche7. Nach der Konditionierung des Furniers bei 21 °C und 50% RH für mindestens einen Tag einen Streifen von 20 mm x 117 mm schneiden. Tragen Sie in der Regel 5 mg Klebstoff gleichmäßig auf 5 mm des Endes eines Holzstreifens auf. Bei 120 °C erhitzten Platten wird das beschichtete Band mit einem anderen Band mit einer Überlappung von 5 mm für 2 min im ABES bei geschlossenen Platten zu einer Lap-Scherprobe verklebt. Nach dem Entfernen der Lap-Shear-Proben aus der ABES-Einheit werden sie über Nacht konditioniert, bevor die ABES-Einheit zur Prüfung der Festigkeit verwendet wird (die Hälfte unter Umgebungsbedingungen und die Hälfte nach dem Eintauchen der Proben in Wasser). Für eine Messung der Haftfestigkeit muss das Versagen im Klebebereich auftreten. Vollständige Details zur Gerätespezifikation sind in der ASTM-Norm11 enthalten.
Das Verfahren ist am nützlichsten für die Bewertung der Festigkeitsentwicklung von Holzklebstoffen in Abhängigkeit von Temperatur und Zeit. Es ist weniger nützlich für Holzklebstoffe, die bei Raumtemperatur aushärten, wie EPI und PUR, da sie keine Wärme für die Verbindung mit Holz benötigen. Grundierungen für Holzklebstoffe, wie HMR, können getestet werden, aber sie werden meist mit Raumtemperaturklebstoffen verwendet. Muster mit Grundierungen könnten mit Furnierstücken, die in die ABES passen, mit einer separaten Presse bei Raumtemperatur verklebt und in der ABES getestet werden.
Die Bedeutung der kleinflächigen Verklebung, wie in ASTM D-7998-19 beschrieben, besteht darin, dass es sich um eine vorläufige Bewertung von Holzklebstoffen handelt, die schnell und mit wenig Aufwand durchgeführt werden kann. Die bestehenden Methoden zur Prüfung von Holzklebstoffen erfordern größere Mengen an Klebstoff und Holz sowie Zeit, um große Platten aus Sperrholz oder Spanplatten zu verkleben, die bei einer bestimmten Temperatur und Feuchtigkeit konditioniert werden müssen, bevor sie von einem professionellen Schreiner in präzise Proben zum Testen zerschnitten werden. Viele Panels müssen hergestellt werden, um verschiedene Variablen zu testen, was mit dem ASTM D-7998-19-Verfahren, ABES, einfacher und schneller durchgeführt werden kann. Es gibt keine andere Testmethode, die kinetische Aushärtungsdaten eines Klebstoffs bestimmen kann.

Holzverklebungen sind der größte Einzelklebstoffmarkt und haben zu einer effizienten Nutzung der Waldressourcen geführt. Viele Jahrhunderte lang wurde Massivholz für die meisten Anwendungen verwendet, mit Ausnahme des Möbelbaus, ohne Testkriterien außer der Haltbarkeit des Produkts. Gebundene Holzprodukte wurden jedoch immer häufiger, beginnend mit Sperrholz und Brettschichtholzbalken, wobei biobasierte Klebstoffe verwendetwurden 1,2. Obwohl diese Produkte zu dieser Zeit zufriedenstellend waren, führte der Ersatz von Soja-, Kasein- und Blutklebstoffen durch formaldehydhaltige synthetische Klebstoffe zu verbesserten Eigenschaften. Die höhere Leistung dieser neuen Klebstoffe führte zu definierten Prüfstandards mit höheren Leistungserwartungen als mit den meisten biobasierten Klebstoffen. Die synthetischen Klebstoffe ermöglichten auch die Verbindung von Partikeln einschließlich Sägemehl zu Spanplatten, Fasern zu Faserplatten mit unterschiedlichen Dichten, Spänen zur Bereitstellung von orientiertem Litzbrett und parallelem Litzenholz, Furnieren zu Sperrholz und Furnierschichtholz sowie keilgezinktem Holz, Brettschichtholz, Brettschichtholz und Holzbalken3. Jedes dieser Produkte hat seine eigenen Prüfkriterien4. Daher kann die Entwicklung eines neuen Klebstoffs viel Formulierungsarbeit und umfangreiche Tests erfordern, um festzustellen, ob ein Potenzial für die Entwicklung einer ausreichenden Festigkeit besteht. Diese zeitaufwändigen Tests und die Komplexität der Holzeigenschaften und Holzverklebungen5 haben die Entwicklung neuer Klebstoffe eingeschränkt. Darüber hinaus können die mechanischen Eigenschaften von Holzklebstoffen unterschiedlich sein, wenn sie zwischen Holzoberflächen ausgehärtet werden, im Gegensatz zu ordentlich6. Durch die Aushärtung in Kontakt mit Holz können Wasser und niedermolekulare Komponenten aus dem Klebstoff entweichen, zusätzlich zu komplexen Interphasen- und chemischen Wechselwirkungen des Klebstoffs mit dem Holz 3,7.
Die Entwicklung des Automated Bonding Evaluation System (ABES) war sehr hilfreich für das Verständnis der Festigkeitsentwicklung von Holzklebstoffen, da es schnell und einfach anzuwenden ist 8,9,10. Das System ist eine integrale Einheit, die Lap-Shear-Proben verbindet und dann die Kraft unter Spannung misst, die erforderlich ist, um die Verbindung zu brechen. Sein Nutzen hat zur Entwicklung der ASTM-Methode D7998-19 geführt, die dieses System11 verwendet. Obwohl dieses System ursprünglich entwickelt wurde, um die Entwicklung der Klebkraft als Funktion von Temperatur und Zeit zu messen, kann es auch die Wärmebeständigkeit von ausgehärteten Klebstoffen sowie die routinemäßige Bewertung der Haftfestigkeit messen. Während es viele Möglichkeiten gibt, die Härtungseigenschaften von Klebstoffen zu messen, von der Gelzeitrheometrie über die dynamische Differenzkalorimetrie bis hin zur dynamisch-mechanischen Analyse und Spektroskopie vieler Arten, misst nur die ABES-Methode die Entwicklung der mechanischen Festigkeit. Dies erfordert ein Gerät, das für Heizung, Kühlung und Zugversuche an Ort und Stelle streng kontrolliert wird11.

Die dritte besteht darin, die Hitzebeständigkeit des Klebstoffs besser zu verstehen, indem die thermische Empfindlichkeit schnell bewertet und zwischen thermischer Erweichung und thermischem Abbau unterschieden wird.

Das Verfahren wurde ausgiebig für die Untersuchung von Proteinklebstoffen im Forest Products Laboratory eingesetzt. Es wurde festgestellt, dass eine Nassklebefestigkeit von weniger als 2 MPa nicht ausreichte, um weitere Holzklebstofftests zu rechtfertigen, während mehr als 3 MPa ein vielversprechendes Ergebnis für weitere Tests war19. Es hat sich als nützlich erwiesen, um die Empfindlichkeit der Holzverarbeitungsbedingungen12,13 nachzuweisen. Weitere Beispiele finden sich in den Frihart-Publikationen7. Die Präzision und Verzerrung der Methode wurde bestimmt (Forschungsbericht RR:D14-1018), wie in ASTM D7998-1911 zusammengefasst.

A Method for Ovarian Follicle Encapsulation and Culture in a Proteolytically Degradable 3 Dimensional System

The ovarian follicle is the functional unit of the ovary that secretes sex hormones and supports oocyte maturation. In vitro follicle techniques provide a tool to model follicle development in order to investigate basic biology, and are further being developed as a technique to preserve fertility in the clinic1-4. Our in vitro culture system employs hydrogels in order to mimic the native ovarian environment by maintaining the 3D follicular architecture, cell-cell interactions and paracrine signaling that direct follicle development 5. Previously, follicles were successfully cultured in alginate, an inert algae-derived polysaccharide that undergoes gelation with calcium ions6-8. Alginate hydrogels formed at a concentration of 0.25% w/v were the most permissive for follicle culture, and retained the highest developmental competence 9. Alginate hydrogels are not degradable, thus an increase in the follicle diameter results in a compressive force on the follicle that can impact follicle growth10. We subsequently developed a culture system based on a fibrin-alginate interpenetrating network (FA-IPN), in which a mixture of fibrin and alginate are gelled simultaneously. This combination provides a dynamic mechanical environment because both components contribute to matrix rigidity initially; however, proteases secreted by the growing follicle degrade fibrin in the matrix leaving only alginate to provide support. With the IPN, the alginate content can be reduced below 0.25%, which is not possible with alginate alone 5. Thus, as the follicle expands, it will experience a reduced compressive force due to the reduced solids content. Herein, we describe an encapsulation method and an in vitro culture system for ovarian follicles within a FA-IPN. The dynamic mechanical environment mimics the natural ovarian environment in which small follicles reside in a rigid cortex and move to a more permissive medulla as they increase in size11. The degradable component may be particularly critical for clinical translation in order to support the greater than 106-fold increase in volume that human follicles normally undergo in vivo .

A new method for ovarian follicle encapsulation in a 3D fibrin-alginate interpenetrating network is described. This system combines structural support with proteolytic degradation to support the development of immature follicles to produce mature oocytes. This method may be applied to culture cell aggregates to maintain cell-cell contacts without limiting expansion.

A Simple Hanging Drop Cell Culture Protocol for Generation of 3D Spheroids

Studies of cell-cell cohesion and cell-substratum adhesion have historically been performed on monolayer cultures adherent to rigid substrates. Cells within a tissue, however, are typically encased within a closely packed tissue mass in which cells establish intimate connections with many near-neighbors and with extracellular matrix components. Accordingly, the chemical milieu and physical forces experienced by cells within a 3D tissue are fundamentally different than those experienced by cells grown in monolayer culture. This has been shown to markedly impact cellular morphology and signaling. Several methods have been devised to generate 3D cell cultures including encapsulation of cells in collagen gels1or in biomaterial scaffolds2. Such methods, while useful, do not recapitulate the intimate direct cell-cell adhesion architecture found in normal tissues. Rather, they more closely approximate culture systems in which single cells are loosely dispersed within a 3D meshwork of ECM products. Here, we describe a simple method in which cells are placed in hanging drop culture and incubated under physiological conditions until they form true 3D spheroids in which cells are in direct contact with each other and with extracellular matrix components. The method requires no specialized equipment and can be adapted to include addition of any biological agent in very small quantities that may be of interest in elucidating effects on cell-cell or cell-ECM interaction. The method can also be used to co-culture two (or more) different cell populations so as to elucidate the role of cell-cell or cell-ECM interactions in specifying spatial relationships between cells. Cell-cell cohesion and cell-ECM adhesion are the cornerstones of studies of embryonic development, tumor-stromal cell interaction in malignant invasion, wound healing, and for applications to tissue engineering. This simple method will provide a means of generating tissue-like cellular aggregates for measurement of biomechanical properties or for molecular and biochemical analysis in a physiologically relevant model.

We describe a simple, rapid method of generating 3D tissue-like spheroids and their potential application to quantify differences in cell-cell interactions.

Measurement of Aggregate Cohesion by Tissue Surface Tensiometry

Rigorous measurement of intercellular binding energy can only be made using methods grounded in thermodynamic principles in systems at equilibrium. We have developed tissue surface tensiometry (TST) specifically to measure the surface free energy of interaction between cells. The biophysical concepts underlying TST have been previously described in detail1,2. The method is based on the observation that mutually cohesive cells, if maintained in shaking culture, will spontaneously assemble into clusters. Over time, these clusters will round up to form spheres. This rounding-up behavior mimics the behavior characteristic of liquid systems. Intercellular binding energy is measured by compressing spherical aggregates between parallel plates in a custom-designed tissue surface tensiometer. The same mathematical equation used to measure the surface tension of a liquid droplet is used to measure surface tension of 3D tissue-like spherical aggregates. The cellular equivalent of liquid surface tension is intercellular binding energy, or more generally, tissue cohesivity. Previous studies from our laboratory have shown that tissue surface tension (1) predicts how two groups of embryonic cells will interact with one another1-5, (2) can strongly influence the ability of tissues to interact with biomaterials6, (3) can be altered not only through direct manipulation of cadherin-based intercellular cohesion7, but also by manipulation of key ECM molecules such as FN8-11 and 4) correlates with invasive potential of lung cancer12, fibrosarcoma13, brain tumor14 and prostate tumor cell lines15. In this article we will describe the apparatus, detail the steps required to generate spheroids, to load the spheroids into the tensiometer chamber, to initiate aggregate compression, and to analyze and validate the tissue surface tension measurements generated.

We describe a method of measuring binding energy, expressible as tissue surface tension, between cells within 3D tissue-like aggregates. Differences in tissue surface tension have been demonstrated to correlate with invasiveness of lung, muscle, and brain tumors, and are fundamental determinants of establishing spatial relationships between different cell types.

Increasing cDNA Yields from Single-cell Quantities of mRNA in Standard Laboratory Reverse Transcriptase Reactions using Acoustic Microstreaming

Correlating gene expression with cell behavior is ideally done at the single-cell level. However, this is not easily achieved because the small amount of labile mRNA present in a single cell (1-5% of 1-50pg total RNA, or 0.01-2.5pg mRNA, per cell 1) mostly degrades before it can be reverse transcribed into a stable cDNA copy. For example, using standard laboratory reagents and hardware, only a small number of genes can be qualitatively assessed per cell 2. One way to increase the efficiency of standard laboratory reverse transcriptase (RT) reactions (i.e. standard reagents in microliter volumes) comprising single-cell amounts of mRNA would be to more rapidly mix the reagents so the mRNA can be converted to cDNA before it degrades. However this is not trivial because at microliter scales liquid flow is laminar, i.e. currently available methods of mixing (i.e. shaking, vortexing and trituration) fail to produce sufficient chaotic motion to effectively mix reagents. To solve this problem, micro-scale mixing techniques have to be used 3,4. A number of microfluidic-based mixing technologies have been developed which successfully increase RT reaction yields 5-8. However, microfluidics technologies require specialized hardware that is relatively expensive and not yet widely available. A cheaper, more convenient solution is desirable. The main objective of this study is to demonstrate how application of a novel "micromixing" technique to standard laboratory RT reactions comprising single-cell quantities of mRNA significantly increases their cDNA yields. We find cDNA yields increase by approximately 10-100-fold, which enables: (1) greater numbers of genes to be analyzed per cell; (2) more quantitative analysis of gene expression; and (3) better detection of low-abundance genes in single cells. The micromixing is based on acoustic microstreaming 9-12, a phenomenon where sound waves propagating around a small obstacle create a mean flow near the obstacle. We have developed an acoustic microstreaming-based device ("micromixer") with a key simplification; acoustic microstreaming can be achieved at audio frequencies by ensuring the system has a liquid-air interface with a small radius of curvature 13. The meniscus of a microliter volume of solution in a tube provides an appropriately small radius of curvature. The use of audio frequencies means that the hardware can be inexpensive and versatile 13, and nucleic acids and other biochemical reagents are not damaged like they can be with standard laboratory sonicators.

We describe a novel method for increasing cDNA yield from single-cell quantities of mRNA in otherwise standard laboratory reverse transcription reactions. The novelty resides in the use of a micromixer, which utilizes the phenomenon of acoustic microstreaming, to mix fluids at microliter scales more effectively than shaking, vortexing or trituration.

Creating Transient Cell Membrane Pores Using a Standard Inkjet Printer

Bioprinting has a wide range of applications and significance, including tissue engineering, direct cell application therapies, and biosensor microfabrication.1-10 Recently, thermal inkjet printing has also been used for gene transfection.8,9 The thermal inkjet printing process was shown to temporarily disrupt the cell membranes without affecting cell viability. The transient pores in the membrane can be used to introduce molecules, which would otherwise be too large to pass through the membrane, into the cell cytoplasm.8,9,11
The application being demonstrated here is the use of thermal inkjet printing for the incorporation of fluorescently labeled g-actin monomers into cells. The advantage of using thermal ink-jet printing to inject molecules into cells is that the technique is relatively benign to cells.8, 12 Cell viability after printing has been shown to be similar to standard cell plating methods1,8. In addition, inkjet printing can process thousands of cells in minutes, which is much faster than manual microinjection. The pores created by printing have been shown to close within about two hours. However, there is a limit to the size of the pore created (~10 nm) with this printing technique, which limits the technique to injecting cells with small proteins and/or particles. 8,9,11
A standard HP DeskJet 500 printer was modified to allow for cell printing.3, 5, 8 The cover of the printer was removed and the paper feed mechanism was bypassed using a mechanical lever. A stage was created to allow for placement of microscope slides and coverslips directly under the print head. Ink cartridges were opened, the ink was removed and they were cleaned prior to use with cells. The printing pattern was created using standard drawing software, which then controlled the printer through a simple print command. 3T3 fibroblasts were grown to confluence, trypsinized, and then resuspended into phosphate buffered saline with soluble fluorescently labeled g-actin monomers. The cell suspension was pipetted into the ink cartridge and lines of cells were printed onto glass microscope cover slips. The live cells were imaged using fluorescence microscopy and actin was found throughout the cytoplasm. Incorporation of fluorescent actin into the cell allows for imaging of short-time cytoskeletal dynamics and is useful for a wide range of applications.13-15

There is a stated Army need for a field-portable, non-steam sterilizer technology that can be used by Forward Surgical Teams, Dental Companies, Veterinary Service Support Detachments, Combat Support Hospitals, and Area Medical Laboratories to sterilize surgical instruments and to sterilize pathological specimens prior to disposal in operating rooms, emergency treatment areas, and intensive care units. The following ensemble of novel, &#8216;clean and green&#8217; chlorine dioxide technologies are versatile and flexible to adapt to meet a number of critical military needs for decontamination6,15. Specifically, the Portable Chemical Sterilizer (PCS) was invented to meet urgent battlefield needs and close critical capability gaps for energy-independence, lightweight portability, rapid mobility, and rugged durability in high intensity forward deployments3. As a revolutionary technological breakthrough in surgical sterilization technology, the PCS is a Modern Field Autoclave that relies on on-site, point-of-use, at-will generation of chlorine dioxide instead of steam. Two (2) PCS units sterilize 4 surgical trays in 1 hr, which is the equivalent throughput of one large steam autoclave (nicknamed &#8220;Bertha&#8221; in deployments because of its cumbersome size, bulky dimensions, and weight). However, the PCS operates using 100% less electricity (0 vs. 9 kW) and 98% less water (10 vs. 640 oz.), significantly reduces weight by 95% (20 vs. 450 lbs, a 4-man lift) and cube by 96% (2.1 vs. 60.2 ft3), and virtually eliminates the difficult challenges in forward deployments of repairs and maintaining reliable operation, lifting and transporting, and electrical power required for steam autoclaves.

Fabrication of Carbon Nanotube High-Frequency Nanoelectronic Biosensor for Sensing in High Ionic Strength Solutions

The unique electronic properties and high surface-to-volume ratios of single-walled carbon nanotubes (SWNT) and semiconductor nanowires (NW) 1-4 make them good candidates for high sensitivity biosensors. When a charged molecule binds to such a sensor surface, it alters the carrier density5 in the sensor, resulting in changes in its DC conductance. However, in an ionic solution a charged surface also attracts counter-ions from the solution, forming an electrical double layer (EDL). This EDL effectively screens off the charge, and in physiologically relevant conditions ~100 millimolar (mM), the characteristic charge screening length (Debye length) is less than a nanometer (nm). Thus, in high ionic strength solutions, charge based (DC) detection is fundamentally impeded6-8.
We overcome charge screening effects by detecting molecular dipoles rather than charges at high frequency, by operating carbon nanotube field effect transistors as high frequency mixers9-11. At high frequencies, the AC drive force can no longer overcome the solution drag and the ions in solution do not have sufficient time to form the EDL. Further, frequency mixing technique allows us to operate at frequencies high enough to overcome ionic screening, and yet detect the sensing signals at lower frequencies11-12. Also, the high transconductance of SWNT transistors provides an internal gain for the sensing signal, which obviates the need for external signal amplifier.
Here, we describe the protocol to (a) fabricate SWNT transistors, (b) functionalize biomolecules to the nanotube13, (c) design and stamp a poly-dimethylsiloxane (PDMS) micro-fluidic chamber14 onto the device, and (d) carry out high frequency sensing in different ionic strength solutions11.

We describe the device fabrication and measurement protocol for carbon nanotube based high frequency biosensors. The high frequency sensing technique mitigates the fundamental ionic (Debye) screening effect and allows nanotube biosensor to be operated in high ionic strength solutions where conventional electronic biosensors fail. Our technology provides a unique platform for point-of-care (POC) electronic biosensors operating in physiologically relevant conditions.

Methods for Characterizing the Co-development of Biofilm and Habitat Heterogeneity

Biofilms are surface-attached microbial communities that have complex structures and produce significant spatial heterogeneities. Biofilm development is strongly regulated by the surrounding flow and nutritional environment. Biofilm growth also increases the heterogeneity of the local microenvironment by generating complex flow fields and solute transport patterns. To investigate the development of heterogeneity in biofilms and interactions between biofilms and their local micro-habitat, we grew mono-species biofilms of Pseudomonas aeruginosa and dual-species biofilms of P. aeruginosa and Escherichia coli under nutritional gradients in a microfluidic flow cell. We provide detailed protocols for creating nutrient gradients within the flow cell and for growing and visualizing biofilm development under these conditions. We also present protocols for a series of optical methods to quantify spatial patterns in biofilm structure, flow distributions over biofilms, and mass transport around and within biofilm colonies. These methods support comprehensive investigations of the co-development of biofilm and habitat heterogeneity.

Biofilms have complex interactions with their surrounding environment. To comprehensively investigate biofilm-environment interactions, we present here a series of methods to create heterogeneous chemical environment for biofilm development, to quantify local flow velocity, and to analyze mass transport in and around biofilm colonies.

Construction of Defined Human Engineered Cardiac Tissues to Study Mechanisms of Cardiac Cell Therapy

Human cardiac tissue engineering can fundamentally impact therapeutic discovery through the development of new species-specific screening systems that replicate the biofidelity of three-dimensional native human myocardium, while also enabling a controlled level of biological complexity, and allowing non-destructive longitudinal monitoring of tissue contractile function. Initially, human engineered cardiac tissues (hECT) were created using the entire cell population obtained from directed differentiation of human pluripotent stem cells, which typically yielded less than 50% cardiomyocytes. However, to create reliable predictive models of human myocardium, and to elucidate mechanisms of heterocellular interaction, it is essential to accurately control the biological composition in engineered tissues. 
To address this limitation, we utilize live cell sorting for the cardiac surface marker SIRP&#945; and the fibroblast marker CD90 to create tissues containing a 3:1 ratio of these cell types, respectively, that are then mixed together and added to a collagen-based matrix solution. Resulting hECTs are, thus, completely defined in both their cellular and extracellular matrix composition.
Here we describe the construction of defined hECTs as a model system to understand mechanisms of cell-cell interactions in cell therapies, using an example of human bone marrow-derived mesenchymal stem cells (hMSC) that are currently being used in human clinical trials. The defined tissue composition is imperative to understand how the hMSCs may be interacting with the endogenous cardiac cell types to enhance tissue function. A bioreactor system is also described that simultaneously cultures six hECTs in parallel, permitting more efficient use of the cells after sorting.

This manuscript describes the creation of defined engineered cardiac tissues using surface marker expression and cell sorting. The defined tissues can then be used in a multi-tissue bioreactor to investigate mechanisms of cardiac cell therapy in order to provide a functional, yet controlled, model system of the human heart.

Development of an In Vitro Ocular Platform to Test Contact Lenses

Currently, in vitro evaluations of contact lenses (CLs) for drug delivery are typically performed in large volume vials,1-6 which fail to mimic physiological tear volumes.7 The traditional model also lacks the natural tear flow component and the blinking reflex, both of which are defining factors of the ocular environment. The development of a novel model is described in this study, which consists of a unique 2-piece design, eyeball and eyelid piece, capable of mimicking physiological tear volume. The models are created from 3-D printed molds (Polytetrafluoroethylene or Teflon molds), which can be used to generate eye models from various polymers, such as polydimethylsiloxane (PDMS) and agar. Further modifications to the eye pieces, such as the integration of an explanted human or animal cornea or human corneal construct, will permit for more complex in vitro ocular studies. A commercial microfluidic syringe pump is integrated with the platform to emulate physiological tear secretion. Air exposure and mechanical wear are achieved using two mechanical actuators, of which one moves the eyelid piece laterally, and the other moves the eyeballeyepiece circularly. The model has been used to evaluate CLs for drug delivery and deposition of tear components on CLs.

Development of an In Vitro Ocular Platform to Test Contact Lenses

Current in vitro models for evaluating contact lenses (CLs) and other eye-related applications are severely limited. The presented ocular platform simulates physiological tear flow, tear volume, air exposure and mechanical wear. This system is highly versatile and can be applied to various in vitro analyses with CLs.
