Alle tierische Verfahren wurden überprüft und genehmigt durch die institutionelle und Animal Care Committee von der University of South Alabama und wurden in Übereinstimmung mit allen Bundes-, Landes- und lokalen Vorschriften durchgeführt. Primärkulturen von Ratte pulmonalen mikrovaskuläre Endothelzellen (PMVECs) wurden von der Zelle Kultur Core Facility der University of South Alabama Center für Lunge Biologie erhalten. Zellen wurden mit der zuvor beschriebenen Verfahren16zubereitet.
1. Generation von zytotoxischen Überstände
Hinweis: Hier verwenden wir zwei verschiedene Stämme von p. Aeruginosa: PA103, die verfügt über eine intakte Typ III-Sekretion in der Lage, die Exoenzymes ExoU und ExoT auf Zielzellen zu übertragen, während der Infektion und ∆PcrV, die einen Typ-III-Sekretion System fehlt und ist nicht in der Lage Exoenzymes auf Zielzellen nach Impfung zu übertragen.
<ol><li>Bakterien auf Vogel-Bonner (VB) Agar-Platten17 Streifen und wachsen über Nacht bei 37 ° C.<ol>
<li>Um Platten vorzubereiten, machen die folgenden Stammlösung (Vogel-Bonner Salze; 10 X Lager): 2 g MgSO4, 20 g Zitronensäure (freie Säure), 100 g K2PO4und 35 g NaH2PO4zu messen. 1 L und Autoklaven für 15 min mit langsamen Auspuff DdH2O hinzufügen.</li>
		<li>Platzieren Sie 7,5 g Agar in 450 mL DdH2O und Autoklaven wie oben beschrieben.</li>
		<li>Legen Sie nach Autoklavieren beide Lösungen in ein Wasserbad von 50 ° C abkühlen lassen.</li>
		<li>50 mL der 10 X VB Salz Vorratslösung zu Agar hinzufügen.</li>
		<li>400 µg/mL (für die Stämme von p. Aeruginosa verwendet wird) Carbenicillin hinzu und mischen Sie gut durch Schütteln der Flasche.</li>

	<li>PMVEC Reinheit überprüfen, indem Sie positive Färbung mit fluoreszierenden Griffonia Simplicifolia Lektine und negative Färbung mit fluoreszierenden Helix Pomatia Lektin (ein Marker für arterielle Endothelzellen). Aliquoten Zellen und Einfrieren in flüssigem Stickstoff.</li>
	<li>Pflegen Sie für Experimente Zellen in Dulbeccos geändert Eagle Medium (DMEM) ergänzt mit 10 % fötalen Rinderserum und wachsen Sie Zellen in einem 37 ° C Inkubator mit 5 % CO2. Verwenden Sie Zellen bei Passagen 5-15.<ol>
<li>Für die Erzeugung von zytotoxischen Überstände Platte 2 x 106 Zellen in einzelne 150 cm Gerichte und wachsen für 3-4 Tage bis Zusammenfluss erreicht ist. Verwenden Sie zwei Platten pro Experiment (siehe unten).</li>
		<li>Für die Analyse von zytotoxischen Überstände Platte 1 x 105 Zellen in einzelnen Vertiefungen eines 6-gut austeilen und für vier Tage, bis Zusammenfluss erreicht wird wachsen.</li>
	</ol>
</li>
	<li>Um überstand zu produzieren, eines der zwei 150 cm Gerichte des PMVECs aus Schritt 1.3.1 mit Phosphat-gepufferte Kochsalzlösung waschen, trypsinize und dann zählen die Zellen (wir verwenden einen automatisierte Zelle Zähler und Folgen des Herstellers Protokoll; sehen Sie Tabelle der Materialien). Das andere Gericht des PMVECs mit Hank es ausgewogen Salz Lösung (HBSS) waschen und dann mit beiden Stamm von p. Aeruginosa bei einer Vielzahl von Infektionen (MOI) von 20:1 zu infizieren. Dies wird folgendermaßen erreicht:<ol>
<li>Für p. Aeruginosa, OD540 0,25 = 2 x 108 KBE/mL. Zur Vorbereitung dieser Verdünnung fügen Sie 1 mL HBSS eine 1 mL-Einweg-Küvette hinzu. Kratzen Sie genug Bakterien aus der Platte, die in Schritt 1.1 vorbereitet wurde, bis ein OD-540 von 0,25 erreicht ist, wenn mit einem Spektralphotometer gemessen. Im nächste Schritt wird eine Berechnung der wieviel Bakterien benötigt werden bieten.</li>
		<li>Sobald die Anzahl der PMVECs in der Kulturschale bestimmt ist (Schritt 1.4 oben), bestimmen die entsprechende Anzahl von Bakterien, die zweite, nicht trypsiniert Kulturschale mit der PMVECs hinzugefügt werden soll. Beispielsweise, wenn feststeht, dass der Kulturschale PMVECs 1,3 x 107 Zellen enthält, dann bereiten Sie 1,3 x 107 Zellen X 20 Bakterien/Zelle X 1 mL/2 x 108 KBE = 1,3 mL Bakterien.</li>
		<li>Verdünnen Sie die 1,3 mL Bakterien in HBSS zu einem Endvolumen von 20 mL, so dass die gesamte Oberfläche des einen 150 cm Schüssel kann mit Flüssigkeit bedeckt sein, dann fügen Sie die verdünnten Bakterien auf die Platte des PMVECs.</li>
		<li>Die PMVECs beimpft mit den Bakterien bei 37 ° C in einem 5 % CO2 Inkubator für 4-5 Std. bis Lücken zu sehen in der Zelle monomolekularen Film bilden, wenn die Platte mikroskopisch beobachtet wird (siehe Abbildung 1 für die richtige Ebene der Spaltbildung) inkubieren.</li>
		<li>Den überstand zu sammeln, dann Zentrifugieren für 10 min bei 2.000 x g in einer Tischplatte Zentrifuge, zelltrümmer zu entfernen. Hinweis: Jede Tischplatte Zentrifuge in der Lage, erzeugen ausreichend g zwingen, pellet-unlysed Zellen und großzellige Fragmente können für diesen Schritt verwendet werden.</li>
		<li>Gießen Sie den überstand in eine Spritze, die einen 0,2 µm-Filter angebracht zu seinem Ende, dann den Überstand durch den Filter zu entfernen Bakterien übergeben hat.</li>
		<li>Verwenden Sie ein Aliquot des sterilen Überstands Zytotoxizität zu testen (siehe 2.1) und den Rest des Überstands bei-80 ° C zur späteren Verwendung einfrieren.</li>

</ol>2. Analyse der zytotoxischen Überstände
<ol><li>Zytotoxizität Assay<ol>
<li>PMVECs in 6-Wohlen Teller bis Zusammenfluss wachsen. Wash-Brunnen hinzufügen, einmal mit HBSS, dann 1,5 mL Filter sterilisiert überstand (gesammelt von entweder geimpft PA103 oder ∆PcrV Zellen) individuelle Vertiefungen. Geben Sie sterile HBSS in einem anderen als Negativkontrolle.</li>
		<li>Legen Sie die Platte in den CO-2 -Inkubator für 21-24 h, dann Zelle Tötung/Zytotoxizität zu beobachten (siehe nächster Schritt). Überstände, die Zytotoxizität für weitere Experimente zeigen, und verwerfen Sie die Zelle Tötung nicht angezeigt werden.</li>
		<li>Quantifizierung der Zelle Tötung<ol>
<li>Maßnahme Laktat-Dehydrogenase (LDH) Entlassung aus abgestorbenen Zellen, die mit im Handel erhältlichen LDH-Assay Kits und des Herstellers empfohlenen Verfahren.</li>
			<li>Alternativ zu quantifizieren Zelle Tötung mikroskopisch mit ImageJ-Software. Dafür, mikroskopische Aufnahmen und Bereichen der Kulturschale mit intakten Zellen aufnehmen, dann quantifizieren Regionen fehlen Zellen (indikativ für Zelltod in einem monomolekularen Film) mit einem benutzerdefinierten ImageJ (National Institutes of Health)-Makro (siehe zusätzliche Codierung Datei ). Falls gewünscht, führen Sie das Makro Schritte manuell wie folgt:<ol>
<li>Bilder (RGB oder Tiff-Format) in das Makro eingegeben und Kontrast auf 15 % gesättigt Pixel. Doppelte Bilder Kontrast angepasst und führen Sie den Befehl "Subtrahieren Hintergrund", um ein kontrastreiches Bild der Zelle und Lücke in das Blickfeld zu erhalten.</li>
				<li>Dieses Bild aus dem Originalbild abziehen und das resultierende Bild mit dem Originalbild mit dem bildrechner "Und"-Funktion kombinieren. Konvertieren Sie das resultierende Bild in schwarz (Lücken) und weiß (Zellen) Maske mit der Schwelle-Funktion (mindestens 0, maximal 5), und verwenden Sie die Funktion "Binär untergraben" Lärm aus dem Bild entfernen.</li>
				<li>Messen Sie das Verhältnis von Schwarz bis weiße Pixel innerhalb des resultierenden Bildes mit "Bereich Bruchteil" Messung. Plotten und gebrochene Bereiche für jeden Zeitpunkt der Behandlung als Prozent der maximalen Spaltbereich äußern.</li>
				<li>Berechnen Sie Mittel zu und vergleichen Sie mit One-Way ANOVA mit Tukey post-hoc-Test, mit p -Werte weniger als 0,05 als bedeutende.</li>

		<li>Verwenden Sie Immunoblot Analyse auf um das Vorhandensein von Amyloid, Oligomere Tau und Aβ in Kultur Überstände zu etablieren. Hier werden einfache in-vitro- Methoden beschrieben, die Demonstration der Generation von zytotoxischen amyloiden während der Infektion mit einer Lungenentzündung verursachen Organismus erlauben. Diese Methoden umfassen eine Zelle Kultur Zytotoxizität Assay, Immunoblotting, Quantifizierung der Zelle Tötung mit ein neues mikroskopische Verfahren und ThT Bindung. Analysen der Zytostatika haben gezeigt, dass sie Amyloid in der Natur (Abbildung 2 und Abbildung 4 sind) und Merkmale einer Prion-12 weisen. Die Generation der zytotoxischen Prion-Moleküle während einer Lungenentzündung bietet eine mögliche Erklärung für die erhöhten Sterberaten bei Lungenentzündung Überlebenden nach ihrer Entlassung Krankenhaus beobachtet. Experimente im Gange sind diese Möglichkeit testen.
Das wichtigste Ereignis für alle beschriebenen Studien ist die Generation der zytotoxischen überstand und zwei wichtige Variablen berücksichtigt werden müssen. Erstens in der beschriebenen Experimente Ratte PMVECs für beide die Infektion Schritt und die Zytotoxizität Assay dienten. Ob andere Zelltypen zur Herstellung von Zellgiften verwendet werden könnte ist nicht im Detail untersucht, und es ist möglich, dass andere Zellen zytotoxischen Überstände nach der Behandlung mit verschiedenen Stämme von p. Aeruginosagenerieren können. Wie gezeigt, die ZELLGIFTE sind Amyloid in der Natur und beinhalten Oligomere Tau und Aβ. Es wäre zu erwarten, dass andere Zelltypen, die Tau und/oder Beta-Amyloid-Protein Ausdrücken, zytotoxine nach bakteriellen Impfung zu generieren, obwohl diese Möglichkeit noch um zu prüfenden induziert werden können. Die beschriebenen Verfahren könnte für die Prüfung verwendet werden, ob andere Zelltypen induziert werden können, um zytotoxische amyloide nach verschiedenen Beleidigungen zu produzieren.
Die zweite wichtige Variable bei der Erzeugung von zytotoxischen Überstände ist die Dauer der Behandlung mit Bakterien. Es ist wichtig, dass man die Inkubation von die PMVECs Fortschritte bis Lücken in der Monolage gesehen werden können. Die Identifizierung von Lücken in der Zelle Monolage ist eine Angabe Exoenzymes durch Pseudomonas-Bakterien in den Host Zelle Zytoplasma injiziert haben und die Exoenzymes aktiv, wodurch Retraktion der Zellrahmen (Abbildung 1) sind. Unsere Erfahrung ist, dass der Zeitpunkt der Rücknahme der Zellrahmen ein zuverlässiger Indikator für Zytotoxin Freisetzung der Überstand. Es ist verlockend zu spekulieren, dass die beiden Ereignisse verbunden sind, wie die zytotoxischen amyloiden freigebend Oligomere Tau gehört und Störung der Tau-Aktivität führt zu Mikrotubuli Depolymerisation die Zelle Formänderungen beiträgt. In Experimenten wo Behandlungen weniger als der Betrag der Zeitaufwand für die Spaltbildung induzieren gewesen, hat kleine reife zytotoxischen Amyloid nachgewiesen.
Die Zytotoxizität Assay beschrieben in diesem Manuskript ist einfach und zuverlässig. Zelltod kann leicht durch mikroskopische Untersuchung beobachtet und quantifiziert direkt von mikroskopischen Aufnahmen. Die hier beschriebenen Methoden wurden ausschließlich für die Bewertung Zytotoxin Produktion nach der Ratte pulmonalen endothelial Zellen mit p. AeruginosaInfektion verwendet. Es scheint wahrscheinlich, dass die Verfahren zu beurteilen, ob zytotoxischen amyloide nach Infektion mit anderen Lungenentzündung verursachen Agenten, einschließlich der beiden nosokomiale-assoziierten produziert werden leicht angepasst werden könnten Bakterien und Bakterien und Viren verantwortlich für ambulant erworbenen Arten von Lungenentzündung. Darüber hinaus wurden die gemeldeten Verfahren nur zur zytotoxischen Amyloid Generation während der Infektion von kultivierten Zellen zu untersuchen. Zellkultur ist ein künstliches System, und die Anweisungen hier am ehesten an die Generation der ZELLGIFTE während der pulmonale Infektion von Tiermodellen und menschlichen Patienten beurteilen angepasst werden konnte. Sammlung von biologischen Flüssigkeiten, wie z. B. alvéolaire Lavage von infizierten Tieren oder Patienten könnte verwendet werden, in die gleichen Arten von Assays berichtet hier zu beurteilen, ob zytotoxische amyloiden während Infektionsprozessen in-vivo generiert werden. Natürlich, die Dauer der Inkubation mit Bakterien können ganz anders sein, wenn in Tiere impfen, und alvéolaire Lavage müssten mehrere Zeitpunkt ultimative Behandlungsdauer bestimmen gesammelt werden.
Immunoblot-Analysen haben gezeigt, dass amyloide in die Überstände, einschließlich Oligomere Tau und Aβ (Abbildung 3) vorhanden sind. Obwohl die Immunoblot-Verfahren standard sind, gibt es ein wichtiger Schritt, der vor Einleitung Immunoblot Analysen durchgeführt werden müssen. Insbesondere ist es wichtig, dass die Überstände konzentrierten mindestens um das Zehnfache vor Immunoblotting sein als der Betrag von Amyloid in der UN-konzentriert überstand unterhalb der Nachweisgrenze von Immunoblot Analyse. Wie hier berichtet, Zentrifugation mit Röhren mit einem Filter ausgestattet ist die Methode, die routinemäßig verwendet für diese Konzentration Schritt, aber es scheint vernünftig, dass andere Methoden, z.B. TCA Niederschlag, ebenso nützlich für diesen Schritt wäre.
Die Quantifizierung der Zelle Tötung mit Bildern stellt einen wichtigen Fortschritt dar. Derzeit Vertrauen verfügbare Verfahren zur Quantifizierung der Zelltod auf den Kauf von ziemlich teuer-Kits, die Messung der LDH-Freisetzung aus abgestorbenen Zellen zu ermöglichen. Der hier beschriebene Test beinhaltet keine Kosten ImageJ Programme kostenlos von der NIH-Website heruntergeladen werden. Die einfache Anpassung von ImageJ-Programme kann ziemlich genaue Messung der Zelle Tötung. Allerdings lohnt es sich, darauf hinzuweisen, dass ein paar kleinere Artefakte sind bei Durchführung des genannten Protokolls anzutreffen. Das Hauptproblem ist, dass zellfragmente wie fokale Kontaktflächen der Kulturschale als Zellen sterben verbunden bleiben können. Diese verbleibende zellfragmente können von der Kamera erkannt werden und geben einen künstlichen Hinweis, den Zellen in der Schale bleiben. Als solches ist es selten, dass ein Wert von 100 % Zelle Tötung mit diesem Verfahren gewonnen wird.
Eine Schwäche des gemeldeten Assay Systems ist, dass der Zytotoxin amyloiden bei der Vorbereitung nie vollständig definiert wurden. Als solche ist die Quantifizierung schwierig. ELISA-Assays lässt sich total anders amyloiden bieten ohne detaillierte Angaben über die einzelnen Arten in jeder Vorbereitung. Bis genauere Tests wie Masse-Spec oder in Vitro synthetisiert Amyloid Oligomere, entwickelt werden können, das beste, das was man, im Moment tun kann ist, starten Sie jedes Experiment mit der gleichen Anzahl von Zellen, die gleiche Dauer inkubieren und stellen Sie sicher, dass Zytotoxizität tritt zum gleichen Preis wie die bisherigen Vorbereitungen, die verwendet wurden. Variation von irgendwelchen der etablierten Normen führt zusätzliche Unsicherheit.
Zusammenfassend lässt sich sagen werden einfache in-vitro- Methoden für den Nachweis der Produktion der giftigen Amyloid Moleküle durch pulmonale Endothelzellen nach Infektion mit einer Lungenentzündung verursachen Agent beschrieben. Die Assays sind zuverlässig und wahrscheinlich angepasst für andere Organismen und für den Einsatz mit biologischen Flüssigkeiten von infizierten Tieren und menschlichen Patienten erhalten werden können. Diese Methoden werden für Studien untersuchen die langfristigen Folgen einer Lungenentzündung und die Etablierung der Mechanismen, die Ende-Organversagen bei Patienten nach Krankenhaus entlassen.

Patienten, die eine Lungenentzündung zu überleben haben erhöhte Sterberaten in den Monaten nach Krankenhaus Entlastung1,2,3,4,5,6. In den meisten Fällen tritt der Tod durch irgendeine Art von Ende-Organversagen einschließlich Nieren-, Lungen-, Herz-, oder Leber Veranstaltungen sowie Schlaganfall5,6. Der Grund für die erhöhte Sterblichkeit bei dieser Patientengruppe wurde nie eingerichtet.
Als wird entweder ambulant erworbener Pneumonie eingestuft oder nosokomiale (nosokomiale) und Agenten, die Pneumonie verursachen können auch Bakterien, Viren, Pilze und Chemikalien. Eine der wichtigsten Ursachen der nosokomialen Pneumonie ist das Bakterium Pseudomonas Aeruginosa. P. Aeruginosa ist ein gramnegatives Organismus, der eine Typ-III-Sekretion-System verwendet, um verschiedene Effektor-Molekülen, genannt Exoenzymes, direkt in das Zytoplasma der Ziel-Zellen7,8übertragen. Während der Infektion von pulmonalen endothelial Zellen gezielt die Exoenzymes verschiedene intrazelluläre Proteine, einschließlich eine endotheliale Form der Mikrotubuli-assoziierten Protein Tau9,10,11,12 , was zu endotheliale Barriere Aufschlüsselung, was zu schweren Lungenödem, verringerte Lungenfunktion und oft zum Tod.
Wie bereits erwähnt, haben Patienten, die überleben die ersten Lungenentzündung Sterberaten in den ersten 12 Monaten nach Krankenhaus entlassen erhöht. Ein möglicher Mechanismus für die Erklärung dieses Phänomens ist, dass irgendeine Art von langlebigen Toxin während der anfänglichen Infektion entsteht, die schlechte Langzeitprognose führt. Zwei Beobachtungen unterstützen diese Möglichkeit. Erste, kultivierte pulmonale Endothelzellen, die ursprünglich mit p. Aeruginosa behandelt werden nicht für vermehren sich bis zu einer Woche nach der Bakterien durch Antibiotika13getötet werden. Zweite, langlebigen Prionen und Agenten mit Prion Eigenschaften wurden in verschiedene Krankheiten bei Mensch und Tier, insbesondere Erkrankungen im Zusammenhang mit dem Nervensystem14,15nachgewiesen.
Methoden für die Prüfung der möglichen Produktion von langlebigen Zytostatika in pulmonale Infektion sind nie beschrieben worden. Hier werden eine Reihe von einfachen in-vitro- Tests beschrieben, die für die Untersuchung von Zytotoxin Produktion und Aktivität nach Infektion mit einem gemeinsamen Lungenentzündung verursachen Mittel, p. Aeruginosaverwendet werden. Diese Tests sollten leicht anpassungsfähig zu untersuchen mögliche Zytotoxin Induktion nach Infektion mit anderen Agenten, die Pneumonie verursachen, und die Überstände, die generiert werden sollte auch für die Untersuchung von Auswirkungen der ZELLGIFTE ganz nützlich sein Organe oder Tiere. Schließlich werden die Assays, die hier, am ehesten beschrieben werden anpassungsfähig, tierische und menschliche biologische Flüssigkeiten zur Herstellung von Zellgiften während einer Lungenentzündung zu testen.

Patienten, die überleben, Lungenentzündung erhöht Sterberaten in den Monaten nach Krankenhaus entlassen. Es ist vermutet worden, dass Infektion des Lungen-Gewebes während der Lungenentzündung führt bei der Herstellung von langlebigen ZELLGIFTE, die nachfolgende Ende Organversagen führen kann. Wir haben in-vitro- Tests um die Hypothese zu testen, während pulmonale Infektion entstehen ZELLGIFTE entwickelt. Isolierte Ratte pulmonalen Endothelzellen und das Bakterium Pseudomonas Aeruginosa dienen als Modellsysteme und die Produktion von Cytoxins nach Infektion der Endothelzellen durch Bakterien zeigt mit Zellkulturen, gefolgt von direkte Quantifizierung mit Laktat-Dehydrogenase Assays und eine neuartige mikroskopische Methode ImageJ-Technologie. Amyloid Artder diese ZELLGIFTE wurde durch Thioflavin T Bindung Assays und Immunoblotting und Immunodepletion mit A11 Anti-Amyloid Antikörper nachgewiesen. Weitere Analysen mittels Immunoblotting nachgewiesen, dass Oligomere Tau und Aβ wurden produziert und veröffentlicht von Endothelzellen nach Infektion durch p. Aeruginosa. Diese Methoden sollten leicht anpassbar an Analysen der menschlichen klinischen Proben sein.

Eine einfache in-vitro- Test wurde entwickelt, um auf das Vorhandensein von Zellgiften in Überstände von Zellen infiziert mit dem Bakterium p. Aeruginosaassay. Grundsätzlich Kulturmedium aus infizierten Zellen ist 4 h nach bakteriellen Zugabe gesammelt, die Bakterien werden durch Filter Sterilisation der Überstand Kultur entfernt und dann der sterile Überstand wird hinzugefügt, um eine neue Population von Zellen. Die Zellen sind dann 21-24 h nach der Zugabe des Überstands beobachtet und Zelle Tötung quantifiziert.
Zugabe von p. Aeruginosa , konfluierende Schichten von PMVECs induziert Bildung von Lücken zwischen den Zellen. In Vivo, Spaltbildung zu Lungenödem und verminderte Lungenfunktion führt. In der beschriebenen Test ist Spaltbildung verwendet, da der Zeitpunkt für das Screening zollrechtlich Zytotoxin Zelle Kultur Überstände und ausreichender Dauer der Inkubation mit PA103 durch das Vorhandensein von Lücken, die in der Zelle monomolekularen Film zu bilden beginnen angezeigt wird, wie in gezeigt Abbildung 1. ΔPcrV Bakterien auslösen kein Zelle Lücken unter diesen Inkubationsbedingungen. Sobald Lücken erkannt werden, ist der Überstand gesammelt, Filter sterilisiert und anschließend zu naive PMVECs Beurteilungder zytotoxische Aktivität hinzugefügt. Die PMVECs in ihren jeweiligen Überstände dann platziert in einem Inkubator für 21-24 h, und dann beobachtet und fotografiert. Wie in Abbildung 2dargestellt, enthalten überstand von PMVECs, die mit p. Aeruginosa Belastungen PA103 infiziert hatte ZELLGIFTE, die Tod von kultivierten Zellen durch 21 h nach der Zugabe des Überstands induziert. Im Gegensatz dazu verzeichneten keine Zelltod Kontrollzellen mit entweder HBSS Medium oder mit überstand gesammelt aus dem ΔPcrV-Stamm der Pseudomonas behandelt. Immunoblot Analyse ergab, dass Amyloid-Moleküle, einschließlich Oligomere Tau und Aβ, entstehen während der Infektion mit dem PA103 Stamm, während keine amyloiden durch die ΔPcrV Belastung, nach Impfung des PMVECs (Abbildung 2) erzeugt werden. Immunodepletion von amyloiden aus der Überstand vor der Zugabe zum kultivierten Zellen12die Rolle der amyloide als ZELLGIFTE in diesem Test bestätigt. Obwohl hier nicht gezeigt, verbraucht Immunodepletion zytotoxische überstand mit A11 Anti-Amyloid Antikörper und T22 Anti-Tau-Oligomer Antikörper vollständig zytotoxische Aktivität von Überstände produziert nach PA103 Infektion (Balczon Et Al. 2017 12 und Balczon Et Al., in Vorbereitung).
Eine neuartige, kostengünstige und einfache Methode zur Quantifizierung der Zelle Tötung wurde entwickelt. Für diesen Test wurde ImageJ-Software angepasst, direkte Messung der Bereiche im mikroskopischen Bereich zuzulassen, die Zellen (indikativ für Zelle Tötung) fehlt. Die Zuverlässigkeit dieser Methode wurde bestätigt durch direkten Vergleich der etablierte Methode der Messzelle zu töten, die LDH-Freisetzung aus den Zellen ist. Wie in Abbildung 3dargestellt, kann ImageJ-Software zur Umwandlung eines mikroskopischen Feldes, die weißen Zellen enthalten und Regionen, die Zellen zu schwarz fehlt. Ein einfaches Verhältnis von weißen Flächen schwarz kann dann verwendet werden, um Zelle Tötung zu messen. Beim Starten mit konfluierende Monolage Zellen sind alle Regionen der mikroskopischen Bereich weiß. Jedoch konnte um 18 h nach der Zugabe der Überstände mit Zellgiften, Lücken in der Monolage nachgewiesen werden. Berechnung von Flächengrößen der Kulturschale frei von Zellen, dann erhöht sich linear bis 36 h nach Zugabe von zytotoxischen überstand. Bei der Messung der LDH-Freisetzung wurden mit LDH Freigabe erkannt wird zunächst um 18 Uhr nach der Zugabe von zytotoxischen überstand identische Ergebnisse erzielt. LDH-Freisetzung erhöht dann sich linear, bis maximal Zelle Tötung bei 36 h nach der Zugabe von überstand gemessen wurde. Kleine Zelle Tod wurde in Zellen mit überstand gesammelt aus ΔPcrV beimpft Zellen (Abbildung 3) behandelt oder in Kulturen mit HBSS behandelt wurden 36 h gemessen.
Die Menge an Amyloid aus Zellen mit Bakterien behandelt entlassen kann durch Messung von ThT Fluoreszenz quantifiziert werden. Bindung von amyloiden, ThT bewirkt eine Konformationsänderungen Verschiebung in das Molekül und eine Erhöhung der Fluoreszenz-Intensität. Dieser Assay Thioflavin T wird hinzugefügt, um eine Küvette und Grundlinie Fluoreszenz wird gemessen. Die Fluoreszenz-Aufnahme wird dann gestoppt, während eine kleine aliquoten Probe hinzugefügt wird. Die Küvette wird dann an die Fluorimeter zurückgegeben und die Änderung ist, dass Fluoreszenzemission aufgenommen wurde. Wie in Abbildung 4dargestellt, überstand von Zellen, die mit PA103 Stamm von p. Aeruginosa enthaltenen Amyloid durch die Zunahme der Fluoreszenzemission inkubiert wurden gesammelt. Im Gegensatz dazu fehlte überstand von PMVECs mit ΔPcrV Stamm beimpft amyloide, wie durch das Fehlen einer erhöhten Fluoreszenz angegeben.

Bacterial biofilms have been associated with a number of different human diseases, but biofilm development has generally been studied on non-living surfaces. In this paper, we describe protocols for forming Pseudomonas aeruginosa biofilms on human airway epithelial cells (CFBE cells) grown in culture. In the first method (termed the Static Co-culture Biofilm Model), P. aeruginosa is incubated with CFBE cells grown as confluent monolayers on standard tissue culture plates. Although the bacterium is quite toxic to epithelial cells, the addition of arginine delays the destruction of the monolayer long enough for biofilms to form on the CFBE cells. The second method (termed the Flow Cell Co-culture Biofilm Model), involves adaptation of a biofilm flow cell apparatus, which is often used in biofilm research, to accommodate a glass coverslip supporting a confluent monolayer of CFBE cells. This monolayer is inoculated with P. aeruginosa and a peristaltic pump then flows fresh medium across the cells. In both systems, bacterial biofilms form within 6-8 hours after inoculation. Visualization of the biofilm is enhanced by the use of P. aeruginosa strains constitutively expressing green fluorescent protein (GFP). The Static and Flow Cell Co-culture Biofilm assays are model systems for early P. aeruginosa infection of the Cystic Fibrosis (CF) lung, and these techniques allow different aspects of P. aeruginosa biofilm formation and virulence to be studied, including biofilm cytotoxicity, measurement of biofilm CFU, and staining and visualizing the biofilm.

Pseudomonas aeruginosa and Saccharomyces cerevisiae Biofilm in Flow Cells

Many microbial cells have the ability to form sessile microbial communities defined as biofilms that have altered physiological and pathological properties compared to free living microorganisms. Biofilms in nature are often difficult to investigate and reside under poorly defined conditions1. Using a transparent substratum it is possible to device a system where simple biofilms can be examined in a non-destructive way in real-time: here we demonstrate the assembly and operation of a flow cell model system, for in vitro 3D studies of microbial biofilms generating high reproducibility under well-defined conditions2,3.
The system consists of a flow cell that serves as growth chamber for the biofilm. The flow cell is supplied with nutrients and oxygen from a medium flask via a peristaltic pump and spent medium is collected in a waste container. This construction of the flow system allows a continuous supply of nutrients and administration of e.g. antibiotics with minimal disturbance of the cells grown in the flow chamber. Moreover, the flow conditions within the flow cell allow studies of biofilm exposed to shear stress. A bubble trapping device confines air bubbles from the tubing which otherwise could disrupt the biofilm structure in the flow cell.
The flow cell system is compatible with Confocal Laser Scanning Microscopy (CLSM) and can thereby provide highly detailed 3D information about developing microbial biofilms. Cells in the biofilm can be labeled with fluorescent probes or proteins compatible with CLSM analysis. This enables online visualization and allows investigation of niches in the developing biofilm. Microbial interrelationship, investigation of antimicrobial agents or the expression of specific genes, are of the many experimental setups that can be investigated in the flow cell system.

Mitochondria participate in many important physiological events, such as energy production, regulation of signaling transduction, and apoptosis. The study of beta cell mitochondrial functional changes during the development of T1D is a novel area of research. Using the mitochondrial membrane potential dye Tetramethyl Rhodamine Methyl Ester (TMRM) and confocal microscopic live cell imaging, we monitored mitochondrial membrane potential over time in the beta cell line NIT-1. For imaging studies, effector AI4 T cells were labeled with the fluorescent nuclear staining dye Picogreen. NIT-1 cells and T cells were co-cultured in chambered coverglass and mounted on the microscope stage equipped with a live cell chamber, controlled at 37&deg;C, with 5% CO2, and humidified. During these experiments images were taken of each cluster every 3 minutes for 400 minutes.
Over a course of 400 minutes, we observed the dissipation of mitochondrial membrane potential in NIT-1 cell clusters where AI4 T cells were attached. In the simultaneous control experiment where NIT-1 cells were co-cultured with MHC mis-matched human lymphocyte Jurkat cells, mitochondrial membrane potential remained intact. This technique can be used to observe real-time changes in mitochondrial membrane potential in cells under attack of cytotoxic lymphocytes, cytokines, or other cytotoxic reagents.

Cell-mediated lymphocytotoxicity (CML) assays can be used to test autoreactive responses and study mechanisms of cell death in vitro. However, using live-cell confocal microscopic imaging techniques with fluorescent dyes, the type and kinetics of cell death as well as the pathways utilized can be studied in greater detail.

Selection of Plasmodium falciparum Parasites for Cytoadhesion to Human Brain Endothelial Cells

Most human malaria deaths are caused by blood-stage Plasmodium falciparum parasites. Cerebral malaria, the most life-threatening complication of the disease, is characterised by an accumulation of Plasmodium falciparum infected red blood cells (iRBC) at pigmented trophozoite stage in the microvasculature of the brain2-4. This microvessel obstruction (sequestration) leads to acidosis, hypoxia and harmful inflammatory cytokines (reviewed in 5). Sequestration is also found in most microvascular tissues of the human body2, 3. The mechanism by which iRBC attach to the blood vessel walls is still poorly understood.
The immortalized Human Brain microvascular Endothelial Cell line (HBEC-5i) has been used as an in vitro model of the blood-brain barrier6. However, Plasmodium falciparum iRBC attach only poorly to HBEC-5i in vitro, unlike the dense sequestration that occurs in cerebral malaria cases. We therefore developed a panning assay to select (enrich) various P. falciparum strains for adhesion to HBEC-5i in order to obtain populations of high-binding parasites, more representative of what occurs in vivo.
A sample of a parasite culture (mixture of iRBC and uninfected RBC) at the pigmented trophozoite stage is washed and incubated on a layer of HBEC-5i grown on a Petri dish. After incubation, the dish is gently washed free from uRBC and unbound iRBC. Fresh uRBC are added to the few iRBC attached to HBEC-5i and incubated overnight. As schizont stage parasites burst, merozoites reinvade RBC and these ring stage parasites are harvested the following day. Parasites are cultured until enough material is obtained (typically 2 to 4 weeks) and a new round of selection can be performed. Depending on the P. falciparum strain, 4 to 7 rounds of selection are needed in order to get a population where most parasites bind to HBEC-5i. The binding phenotype is progressively lost after a few weeks, indicating a switch in variant surface antigen gene expression, thus regular selection on HBEC-5i is required to maintain the phenotype.
In summary, we developed a selection assay rendering P. falciparum parasites a more "cerebral malaria adhesive" phenotype. We were able to select 3 out of 4 P. falciparum strains on HBEC-5i. This assay has also successfully been used to select parasites for binding to human dermal and pulmonary endothelial cells. Importantly, this method can be used to select tissue-specific parasite populations in order to identify candidate parasite ligands for binding to brain endothelium. Moreover, this assay can be used to screen for putative anti-sequestration drugs7.

RNA Isolation of Pseudomonas aeruginosa Colonizing the Murine Gastrointestinal Tract

Pseudomonas aeruginosa (PA) infections result in significant morbidity and mortality in hosts with compromised immune systems, such as patients with leukemia, severe burn wounds, or organ transplants1. In patients at high-risk for developing PA bloodstream infections, the gastrointestinal (GI) tract is the main reservoir for colonization2, but the mechanisms by which PA transitions from an asymptomatic colonizing microbe to an invasive, and often deadly, pathogen are unclear. Previously, we performed in vivo transcription profiling experiments by recovering PA mRNA from bacterial cells residing in the cecums of colonized mice 3 in order to identify changes in bacterial gene expression during alterations to the host&#x2019;s immune status.
As with any transcription profiling experiment, the rate-limiting step is in the isolation of sufficient amounts of high quality mRNA. Given the abundance of enzymes, debris, food residues, and particulate matter in the GI tract, the challenge of RNA isolation is daunting. Here, we present a method for reliable and reproducible isolation of bacterial RNA recovered from the murine GI tract. This method utilizes a well-established murine model of PA GI colonization and neutropenia-induced dissemination4. Once GI colonization with PA is confirmed, mice are euthanized and cecal contents are recovered and flash frozen. RNA is then extracted using a combination of mechanical disruption, boiling, phenol/chloroform extractions, DNase treatment, and affinity chromatography. Quantity and purity are confirmed by spectrophotometry (Nanodrop Technologies) and bioanalyzer (Agilent Technologies) (Fig 1). This method of GI microbial RNA isolation can easily be adapted to other bacteria and fungi as well.

Following Cell-fate in E. coli After Infection by Phage Lambda

The system comprising bacteriophage (phage) lambda and the bacterium E. coli has long served as a paradigm for cell-fate determination1,2. Following the simultaneous infection of the cell by a number of phages, one of two pathways is chosen: lytic (virulent) or lysogenic (dormant)3,4. We recently developed a method for fluorescently labeling individual phages, and were able to examine the post-infection decision in real-time under the microscope, at the level of individual phages and cells5. Here, we describe the full procedure for performing the infection experiments described in our earlier work5. This includes the creation of fluorescent phages, infection of the cells, imaging under the microscope and data analysis. The fluorescent phage is a "hybrid", co-expressing wild- type and YFP-fusion versions of the capsid gpD protein. A crude phage lysate is first obtained by inducing a lysogen of the gpD-EYFP (Enhanced Yellow Fluorescent Protein) phage, harboring a plasmid expressing wild type gpD. A series of purification steps are then performed, followed by DAPI-labeling and imaging under the microscope. This is done in order to verify the uniformity, DNA packaging efficiency, fluorescence signal and structural stability of the phage stock. The initial adsorption of phages to bacteria is performed on ice, then followed by a short incubation at 35&deg;C to trigger viral DNA injection6. The phage/bacteria mixture is then moved to the surface of a thin nutrient agar slab, covered with a coverslip and imaged under an epifluorescence microscope. The post-infection process is followed for 4 hr, at 10 min interval. Multiple stage positions are tracked such that ~100 cell infections can be traced in a single experiment. At each position and time point, images are acquired in the phase-contrast and red and green fluorescent channels. The phase-contrast image is used later for automated cell recognition while the fluorescent channels are used to characterize the infection outcome: production of new fluorescent phages (green) followed by cell lysis, or expression of lysogeny factors (red) followed by resumed cell growth and division. The acquired time-lapse movies are processed using a combination of manual and automated methods. Data analysis results in the identification of infection parameters for each infection event (e.g. number and positions of infecting phages) as well as infection outcome (lysis/lysogeny). Additional parameters can be extracted if desired.

Long Term Chronic Pseudomonas aeruginosa Airway Infection in Mice

A mouse model of chronic airway infection is a key asset in cystic fibrosis (CF) research, although there are a number of concerns regarding the model itself. Early phases of inflammation and infection have been widely studied by using the Pseudomonas aeruginosa agar-beads mouse model, while only few reports have focused on the long-term chronic infection in vivo. The main challenge for long term chronic infection remains the low bacterial burden by P. aeruginosa and the low percentage of infected mice weeks after challenge, indicating that bacterial cells are progressively cleared by the host.
This paper presents a method for obtaining efficient long-term chronic infection in mice. This method is based on the embedding of the P. aeruginosa clinical strains in the agar-beads in vitro, followed by intratracheal instillation in C57Bl/6NCrl mice. Bilateral lung infection is associated with several measurable read-outs including weight loss, mortality, chronic infection, and inflammatory response. The P. aeruginosa RP73 clinical strain was preferred over the PAO1 reference laboratory strain since it resulted in a comparatively lower mortality, more severe lesions, and higher chronic infection. P. aeruginosa colonization may persist in the lung for over three months. Murine lung pathology resembles that of CF patients with advanced chronic pulmonary disease.
This murine model most closely mimics the course of the human disease and can be used both for studies on the pathogenesis and for the evaluation of novel therapies.

In order to study human acute lung injury and pneumonia, it is important to develop animal models to mimic various pathological features of this disease. Here we have developed a mouse lung injury model by intra-tracheal injection of bacteria Pseudomonas aeruginosa (P. aeruginosa or PA). Using this model, we were able to show lung inflammation at the early phase of injury. In addition, alveolar epithelial barrier leakiness was observed by analyzing bronchoalveolar lavage (BAL); and alveolar cell death was observed by Tunel assay using tissue prepared from injured lungs. At a later phase following injury, we observed cell proliferation required for the repair process. The injury was resolved 7 days from the initiation of P. aeruginosa injection. This model mimics the sequential course of lung inflammation, injury and repair during pneumonia. This clinically relevant animal model is suitable for studying pathology, mechanism of repair, following acute lung injury, and also can be used to test potential therapeutic agents for this disease.

Pneumococcus Infection of Primary Human Endothelial Cells in Constant Flow

Interaction of Streptococcus pneumoniae with the surface of endothelial cells is mediated in blood flow via mechanosensitive proteins such as the Von Willebrand Factor (VWF). This glycoprotein changes its molecular conformation in response to shear stress, thereby exposing binding sites for a broad spectrum of host-ligand interactions. In general, culturing of primary endothelial cells under a defined shear flow is known to promote the specific cellular differentiation and the formation of a stable and tightly linked endothelial layer resembling the physiology of the inner lining of a blood vessel. Thus, the functional analysis of interactions between bacterial pathogens and the host vasculature involving mechanosensitive proteins requires the establishment of pump systems that can simulate the physiological flow forces known to affect the surface of vascular cells.
The microfluidic device used in this study enables a continuous and pulseless recirculation of fluids with a defined flow rate. The computer-controlled air-pressure pump system applies a defined shear stress on endothelial cell surfaces by generating a continuous, unidirectional, and controlled medium flow. Morphological changes of the cells and bacterial attachment can be microscopically monitored and quantified in the flow by using special channel slides that are designed for microscopic visualization. In contrast to static cell culture infection, which in general requires a sample fixation prior to immune labeling and microscopic analyses, the microfluidic slides enable both the fluorescence-based detection of proteins, bacteria, and cellular components after sample fixation; serial immunofluorescence staining; and direct fluorescence-based detection in real time. In combination with fluorescent bacteria and specific fluorescence-labeled antibodies, this infection procedure provides an efficient multiple component visualization system for a huge spectrum of scientific applications related to vascular processes.

This study describes the microscopic monitoring of pneumococcus adherence to von Willebrand factor strings produced on the surface of differentiated human primary endothelial cells under shear stress in defined flow conditions. This protocol can be extended to detailed visualization of specific cell structures and quantification of bacteria by applying differential immunostaining procedures.

Culture of Small Colony Variant of Pseudomonas aeruginosa and Quantitation of its Alginate

Pseudomonas aeruginosa, an opportunistic Gram-negative bacterial pathogen, can overproduce an exopolysaccharide alginate resulting in a unique phenotype called mucoidy. Alginate is linked to chronic lung infections resulting in poor prognosis in patients with cystic fibrosis (CF). Understanding the pathways that regulate the production of alginate can aid in the development of novel therapeutic strategies targeting the alginate formation. Another disease-related phenotype is the small colony variant (SCV). SCV is due to the slow growth of bacteria and often associated with increased resistance to antimicrobials. In this paper, we first show a method of culturing a genetically defined form of P. aeruginosa SCV due to pyrimidine biosynthesis mutations. Supplementation of nitrogenous bases, uracil or cytosine, returns the normal growth to these mutants, demonstrating the presence of a salvage pathway that scavenges free bases from the environment. Next, we discuss two methods for the measurement of bacterial alginate. The first method relies on the hydrolysis of the polysaccharide to its uronic acid monomer followed by derivatization with a chromogenic reagent, carbazole, while the second method uses an ELISA based on a commercially available, alginate-specific mAb. Both methods require a standard curve for quantitation. We also show that the immunological method is specific for alginate quantification and may be used for the measurement of alginate in the clinical specimens.
